摘  要:目的探讨c-myc蛋白表达与c-myc基因易位的相关性,旨在寻找适用于临床c-myc蛋白的界定值,以有效判断c-myc基因易位状态。方法采用免疫组化法与荧光原位杂交(fluorescence in situ hybridization,FISH)法检测293例弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)中c-myc蛋白表达水平与c-myc基因易位状态,分析两者的相关性。结果 293例DLBCL中,30例(10.2%)发生c-myc基因易位,其中29例c-myc蛋白阳性细胞百分率>40%,1例c-myc蛋白阳性细胞百分率为40%,而c-myc蛋白阳性细胞百分率<40%者均未发生c-myc基因易位。随着c-myc蛋白表达水平的升高,c-myc基因易位的发生率也随之升高。结论 c-myc蛋白高表达与c-myc基因易位密切相关。c-myc蛋白可用于DLBCL中c-myc基因易位的筛选,建议对c-myc蛋白阳性细胞百分率>40%者行FISH检测以进一步明确有无c-myc基因易位。Purpose To study the correlation between c-myc expression and c-myc translocation, in order to find out an appropriate cut-off level of c-myc expression for estimation of c-myc translocation in clinical practice. Methods In 293 cases of diffuse large B- cell lymphoma (DLBCL), e-myc expression was observed by immunohistochemistry (IHC) and its translocation was detected by fluo- rescence in situ hybridization (FISH). And the correlation between c-myc expression and translocation was subsequently analyzed. Re- sults In 293 cases of DLBCL, c-myc translocation was detected in 30 ( 10. 2% ) of cases. 29 of them were c-myc expression 〉 40% , and the remained one was 40%. While, none of the cases with c-myc expression 〈 40% had c-myc translocation. Furthermore, the higher c-myc expressed, the more c-myc translocation occurred. Conclusion C-myc overexpression is obviously correlated with c-myc translocation. It is useful to detect the level of e-mye expression in DLBCL so that to evaluate whether it had c-myc translocation. It is suggested that it should be further examined c-myc translocation condition by FISH when its level of c-myc expression is higher than 40%.
