摘  要:为弄清发情期大熊猫阴道内可培养真菌的种类及对药物的敏感性,为大熊猫的生殖道合理用药提供参考,对10只发情大熊猫阴道分泌物进行真菌分离培养、形态学鉴定并用真菌ITS序列通用引物对分离真菌进行PCR扩增、序列测定等;对分离到的5株酵母菌选用5种抗真菌药进行药敏试验。结果,从大熊猫阴道分泌物中共分离到30种真菌,包含25种霉菌和5种酵母菌(链状假丝酵母菌、担子菌类酵母菌、季也蒙毕赤氏酵母菌、汉氏德巴利氏酵母菌、皮状丝孢酵母菌)。5种酵母菌均对两性霉素B、制霉菌素敏感;皮状丝孢酵母菌对酮康唑、伊曲康唑中度敏感;汉氏德巴利氏酵母菌对伊曲康唑中度敏感;链状假丝酵母菌对氟康唑中度敏感,对酮康唑、伊曲康唑耐药。研究结果表明,发情期大熊猫阴道可培养真菌种类主要为霉菌(83.33%),酵母菌较少(16.67%);两性霉素B和制霉菌素是临床治疗由本研究中5种酵母菌引起的大熊猫阴道疾病的首选药物。In order to understand the culturable fungi species from the vagina of estrus giant panda and to provide a reference for rational drug use in giant pandas' reproductive tract,we have isolated and cultivated,and identified based on morphology of the fungi from 10 estrus female pandas vaginal secretions samples.PCR amplification and sequence analysis was performed using fungal ITS universal primers,and then the similarity of their sequences in the NCBI BLAST was compared. Selecte 5 kinds of antifungal drugs to do Susceptibility testing by the Danish rosco disc diffusion method for isolated 5 strains of yeast. We had obtained 30 different fungi,including on 25 species of mycete,5 species of yeasts(Candida catenulata,Pseudozyma aphidis,Pichia guil-liermondii,Debaryomyces hansenii,Trichosporon cutaneum).Susceptibility test showed that the 5 species of yeasts all showed the most sensitive for amphotericin B and nystatin. Trichosporon cutaneum is moderate sensitive to ketoconazole and itraconazole. Debaryomyces hansenii is moderate sensitive to itraconazole.Candida catenulata is moderate sensitive to fluconazole and has drug resistance for ketoconazole,itraconazole. The results showed that the giant panda vaginal culturable fungi species mainly included mycete(83.33%),and yeast was less(16.67%). Amphotericin B and nystatin are preferred drug for pandas' vaginal disease caused by the studied 5yeasts in our study in clinical treatment.
