摘 要:目的探讨不同方案紫外线(UV)照射对白血病HL-60细胞增殖的影响,寻找合适的UV照射方法和照射量。方法以HL-60细胞株为研究对象,并设置对照组和UV照射组(简称UV组),对照组为不接受UV照射的HL-60细胞,UV组细胞用波长为254 nm的UV照射,照射量分别为2 m Joules(m J)、6 m J、10 m J和15 m J;将UV组和HL-60对照组细胞置于倒置相差显微镜下观察细胞生长情况,用台盼蓝染色法测定各组细胞的死亡率。结果未接受照射的对照组HL-60细胞增殖未受影响,未出现明显凋亡;用2~10 m J UV照射1次,可对HL-60细胞的增殖产生抑制,细胞部分凋亡;经15m JUV照射1次或2~10 m J照射2次(每次间隔5 d),多数HL-60细胞出现裂解或细胞完全死亡,不能再恢复增殖;10 m J组细胞UV照射1次后第1天开始出现死亡,细胞死亡率在照射后第3天达最高值,随着培养的继续,细胞死亡率逐渐降低,照射后第3天细胞死亡率与其他时间比较差异有统计学意义(P〈0.05)。结论 UV照射对HL-60细胞具有显著的杀伤效果,与高剂量UV照射治疗相比,较低UV照射剂量的重复照射也能杀伤HL-60细胞,诱导HL-60细胞的死亡。Objective To observe the effect of ultraviolet(UV) radiation on the proliferation of acute promyelocytic leukemia(APL) cell line HL-60,and to investigate the appropriate irradiation method and exposure dose of UV. Methods HL-60 cells were taken as the study objects,control group and UV group were set up,the UV group exposure to 2mJoules(mJ),6mJ,10 mJ and 15 mJ of UV,the wavelength of UV was 254 nm,and the control group did not receive UV. The proliferation of HL-60 cells was observed by phase-contrast microscopy,and the cell mortality rate of HL-60 cells was detected by trypan blue staining.Results No significant apoptosis of HL-60 cells was found in the control group; some apoptosis cells were found and the proliferation of HL-60 cells was inhibited while received 2- 10 mJ UV for once time; must cells were lysed or completely die while irradiated by15 mJ UV once time or 2- 10 mJ UV twice time(once every 5 days); after exposured to 10 mJ UV,dead cells was found in the first day,and the mortality rate reach the highest value on the third day after irradiation,which had statistically significant differences in comparison with the other groups(P 〈 0. 05),and the mortality rate decreased a long with culture. Conclusions UV radiation has a significant killing effect on HL-60 cells,as compared with high doses of UV irradiation,repeated exposure to UV of lower doses can also induced apoptosis of HL-60 cells.
