摘 要:目的探讨B7-H4基因的小干扰RNA(small interference RNA,siRNA)对卵巢癌SKOV-3细胞生长及增殖的影响。方法化学合成B7-H4-siRNA,转染SKOV-3细胞,MTT法检测卵巢癌细胞增殖情况,细胞计数法计算细胞倍增时间。结果 B7-H4-siRNA成功转入SKOV-3细胞,B7-H4-siRNA组细胞生长受到明显抑制,细胞倍增时间明显长于未转染组、随机乱码组和转染试剂对照组。结论 B7-H4-siRNA能沉默SKOV-3细胞中B7-H4的表达,抑制SKOV-3细胞的生长增殖,B7-H4可能成为卵巢癌基因治疗的候选靶点。Objective To investigate the effect of silencing B7-H4 expression by small interference RN A( siRN A)on proliferation of ovarian carcinoma cells. Methods Chemical synthesized siRN A targeting B7-H4 was transfected into SKO V-3 cells. The proliferation of SKO V-3 cells was determined by MTTassay and cell doubling time was detected by cell counting method. Results B7-H4-siRN A was successfully transfected into SKO V-3 cells. In the B7-H4-siRN A group,the proliferation of SKO V-3 cells was significantly inhibited,and the doubling time was longer than that in the untransfected group,the Ctrl-siRN A group and the transfection reagent group. Conclusion B7-H4-siRN A can silence B7-H4 expression in SKO V-3 cells,and inhibit the proliferation of SKO V-3 cells. B7-H4 can be regarded as a candidate gene for ovarian carcinoma gene therapy.
