结直肠腺癌CCRK基因表达在分化程度评估中的意义

摘　　要：目的检测不同分化程度结直肠腺癌患者CCRK基因及其蛋白表达水平,探讨CCRK在结直肠腺癌患者分化程度中的作用,为结直肠腺癌分化程度及其预后提供分子生物学依据。方法分别采用RT-PCR、免疫组织化学方法检测234例结直肠腺癌组织中CCRK mRNA和CCRK蛋白质表达水平。结果在234例结直肠腺癌组织中,CCRK的阳性表达者140例,不表达者94例。其中CCRK阳性表达者在不同分化程度的结直肠腺癌患者中CCRK蛋白染色得分值不同(HC=123.765,P<0.0001);而且CCRKmRNA相对表达量在结直肠癌低分化组(3.71±0.41)明显高于中分化组(3.09±0.23)和高分化组(2.14±0.21),差别具有统计学意义。结论CCRK的表达与结直肠腺癌分化程度有关,CCRK蛋白及其CCRKmRNA相对表达量越高,其分化程度越低,恶性程度也越高;检测CCRK蛋白表达和CCRK mRNA水平有助于结直肠腺癌分化程度和预后的判断。Objective To explore the relationship between CCRK gene and protein expression in different differentiation degree types of colorectal adenocarcinoma,which could provide molecular basis for colorectal adenocarcinoma differentiation and prognosis. Methods The expression of mRN A CCRK and CCRK protein in 234 cases of colorectal adenocarcinoma were detected by RT-PCRand immunohistochemistry respectively. Results There were 140 cases of CCRK positive expression in 234 cases of colorectal adenocarcinoma. CCRK protein expression levels and differentiation degrees staining score values had certain relevance in colorectal adenocarcinoma tissue,and the difference had significant significance in each group（ HC= 123. 765,P 〈0. 0001）. CCRKmRN A relative expression amount（ 3. 71 ± 0. 41） in poorly differentiated colorectal adenocarcinoma was significantly higher than that in moderate differentiation group（ 3. 09± 0. 23） and high differentiation group（ 2. 14 ± 0. 21）. The overall difference was statistically significant（ F = 5. 689,P〈 0. 05）,and the difference between any two groups were statistically significant. It shows that there are positive correlation between CCRK mRN A relative expression and differentiation degree in colorectal adenocarcinoma. Conclusion The expression of CCRK was correlated with the differentiate degree in colorectal adenocarcinoma. And the more higher CCRK expression,the more lower the differentiate degree and the higher the degree of malignancy. The detection of CCRK protein expression and mRN A CCRK levels had contributed to the differentiate degree and prognosis of colorectal adenocarcinoma tumors.
