摘  要:目的探讨阿尔茨海默病与老年2型糖尿病患者尿液中组蛋白的变化及其机制。方法对22例正常人、20例T2DM患者和20例AD患者的尿液标本进行多肽的富集,然后采用多肽组学分析技术进行检测并使用Protein Pilot软件(v4.5)对获得的数据进行检索比对分析,从而确定相应蛋白质的种类。结果与正常对照组相比AD组与T2DM组尿液中组蛋白H2B、组蛋白H2A、组蛋白H4显著增加。结论炎症机制可能导致AD与T2DM患者尿液中组蛋白含量增加。Objectives By investigate the level of histone in urine of patients with Alzheimer's disease( AD) or elderly patients with type 2 diabetes mellitus( T2DM) to seek whether there has common pathogenesis between AD and T2 DM. Methods The urine samples obtained from 22 healthy individuals and 20 patients with T2 DM and 20 patients with AD. The protein were analyzed by mass spectrometry after enrichment using weak cationic-exchange magnetic beads. Then the datum were analyzed by protein Pilot( V. 4. 5) to determine the type of the protein. Results Compared with normal control group,the significant increased protein are histone H2 B,histone H2 A and histone H4 in both of AD and T2 DM patients. Conclusions Histone deacetylases and inflammation may have an effect on the high level of histone in urine.
