摘 要:目的:观察高脂饮食所致非酒精性脂肪肝的形成和停止造模后自愈情况。方法:将健康雄性SD大鼠42只,随机分为7组:正常组、模型1、2、3、4、5、6组,每组6只。正常组予标准饲料,模型组均给予高脂饲料,模型组大鼠10周后改为标准鼠料喂养。分别于实验第2、4、6、8、10、14周将模型1—6组依次全部处死,正常组于第14周处死,观察各组非酒精性脂肪肝大鼠模型的血清、肝脏生化指标及组织病理学变化。结果:与正常组相比,大鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆固醇(TC)、甘油三脂(TG)和大鼠肝指数、Lee's指数于造模第4周时开始升高,到第8周达到高峰,于第14周(即停止高脂饲料4周后)有降低趋势。在肝组织切片在病理学和形态学上,造模组大鼠于第2周肝脏开始出现肝细胞脂肪变、气球样变、小叶炎症和门脉炎症,随着造模时间的延长,上述病理改变加重,于造模第6周开始有大鼠肝脏出现纤维化,并呈重度非酒精性脂肪肝病理变化。而第14周(即停止高脂饲料4周后)肝脏自然愈合在组织病理学上不明显。结论:高脂饲料喂养8—10周内可以成功诱导非酒精性脂肪肝大鼠模型,且模型在停止造模4周内较为稳定。Objective:To observe the high fat diet inducing non -alcoholic fatty liver and stop healing situation after modeling. Methods : Forty - two healthy male SD rats were randomly divided into 7 groups : normal group, model 1,2,3,4, 5,6 group,6 rats in each group. Normal group received standard diet. Model groups were fed with high fat diet and after 10 weeks were given the standard feeding. In experiment,the rats in the model groups 1 to 6 were killed in 2nd ,4th ,6th, 8th, 10th, 14th week and the rats of normal group 14 weeks later also were killed. And then observe the changes of bio- chemical indexes and serum,liver tissue pathology in rats. Results : Compared with normal group, the rats' serum glutamie oxaloacetic transaminase (AST) , alanine aminotransferase (ALT) , total cholesterol ( TC ) , glycerin three fat (TG) and liver index and Lee's index began to increase from 4th week and reached the peak in the 8th week, but in 14th week ( namely stop high fat diet for 4 weeks) it had a decreasing tendency. In liver tissue sections' pathology and morphology, the model rats' liver appeared hepatic steatosis, ballooning, lobular inflammation and portal inflammation in 2rid week. With the modeling time prolongs, there was serious pathological change and from 6th week there was liver fibrosis and nonalcoholic fatty liver pathological changes. In 14h week (namely stop with high fat diet for 4 weeks) ,the natural heal- ing in liver histopathology was not obvious. Conclusion: High fat diet 8 - 10 weeks could successfully induce non - alco- holic fatty liver model and the model is stable within 4 weeks after stopping modeling.
