摘 要:目的:探讨黄芪多糖对血管性痴呆大鼠空间记忆和海马NMDAR1表达的影响。方法:选择血管性痴呆大鼠60只为研究对象,分成对照组和观察组,各30只。对照组:尼莫地平灌胃给药。观察组:黄芪多糖灌胃给药。灌胃给药4周后,通过Morris实验比较两组大鼠上台潜伏期、上台总路程、站台象限停留时间、穿越站台次数的水平。比较灌胃时、灌胃后4周两组大鼠Ach、Ach E水平的变化。比较两组大鼠海马CA1区NMDA基因表达。结果:观察组大鼠上台总路程、大鼠上台潜伏期、大鼠站台象限停留时间、都少于对照组,差异均有统计学意义(P<0.01)。观察组大鼠穿越站台的次数多于对照组,差异均有统计学意义(P<0.01)。观察组大鼠的Ach水平低于对照组,而观察组大鼠的Ach E水平则高于对照组,差异均有统计学意义(P<0.01)。观察组大鼠海马CA1区NMDAR1蛋白表达水平都高于对照组,差异均有统计学意义(P<0.01)。结论:黄芪多糖能够改善血管性痴呆大鼠的空间记忆,促进大鼠海马CA1区受损神经元的恢复。Objective: To investigate the effect of astragalus polysaccharide on the spatial memory and hippocampal NMDAR1 expression in rats with vascular dementia. Method : Sixty rats of vascular dementia rats as the research object - were divided into control group and observation group,30 rats in each. Control group:nimodipine by intragastric adminis- tration. The observation group:astragalus polysaccharide by intragastric administration. After intragastric administration for 4 weeks, Morris experiment was used to compare rats' latency, total distance, platform quadrant residence time through and the number of crossing platform in the two groups. Compare Ach and AchE levels in the two groups. Compare NM- DA gene expression in hippocampal CA1 area of rats in the two groups. Results : The total distance and the latency and platform quadrant residence time in the observation group were less than those of the control group and the differences were statistically significant ( P 〈 0.01 ). The number of rats crossing the platform in the observation group than the control group's and the differences were statistically significant ( P 〈 0.01 ). The Ach level in observation group was lower than that in the control group. While the level of Ache in the observation group was higher than the control group's and there was statistically significant difference ( P 〈0.01 ). The expression of NMDAR1 in hippocampal CA1 of ob- servation group was higher than that in control group. The differences were statistically significant ( P 〈 0. 01 ). Conclusion: Astragalus polysaccharide can improve the spatial memory in vascular dementia rats and promote the dam- aged neurons recovery of hippocampal CAI region of rats.
