摘 要:为了解CYP2E1基因单核苷酸多态性在从江香猪中的分布,积累从江香猪药物代谢模型基础资料,应用SNa Pshot分型技术对141份从江香猪样品CYP2E1基因13个错义突变位点进行基因分型,并统计群体遗传学指标。结果显示:rs694867178、rs693643523、rs321622043、rs706465876、rs690804887、rs707424098、rs698846226、rs708577553、rs695879722和rs80932038共10个位点表现为单态,rs793477975、rs691870665和rs80969284共3个位点表现为多态。多态位点的基因型分布频率为:rs793477975(CC 95.74%,CT 4.26%)、rs691870665(AA 9.93%,AG 32.62%,GG 57.45%)、rs80969284(AA 7.8%,AG 33.33%和GG 58.87%);等位基因分布频率为:rs793477975(C 97.87%,T 2.13%)、rs691870665(A 26.24%,G 73.76%)、rs80969284(A 24.47%,G 75.53%)。rs793477975位点多态信息含量、杂合度、香农信息指数都很低,有效等位基因数小,变异小;rs691870665和rs80969284位点多态信息含量、杂合度、香农信息指数都较高,有效等位基因数较大,遗传变异较大。结果提示,从江香猪CYP2E1基因总体上纯合度高,在药物代谢动物模型开发上具有良好基础。In order to figure out the distribution of single nucleotide polymorphisms of CYP2E 1 gene in Congjiang Xiang-pigs, and then accumulate data for drug metabolism model using Congiiang xiang-pigs, 13 missense SNPs of CYP2E1 gene were genotyped by SNaPshot method in 141 Congjiang Xiang-pigs. Moreover, the population genetic indexes were analyzed. The results indicated that the total of 10 loci of rs694867178, rs693643523, rs321622043, rs706465876, rs690804887, rs707424098, rs698846226, rs708577553, rs695879722 and rs80932038 were monomorphic, and the 3 loci of rs793477975, rs691870665 and rs80969284 were polymorphic. In 3 polymo- rphic locis, the frequencies ofgenotype distribution ofCYP2E1 gene were: rs793477975 (CC 95.74% and CT 4.26%), rs691870665 (AA 9.93%, AG 32.62% and GG 57.45%) and rs80969284 (AA 7.8%, AG 33.33% and GG 58.87%). The frequencies of allele distribution were: rs793477975 (C 97.87% and T 2.13%), rs691870665 (A 26.24% and G 73.76%) and rs80969284 (A 24.47% and G 75.53%). In rs793477975 locus, very low polymorphism information content, heterozygosity and Shannon's information index were observed, and there were small effective number of alleles and little genetic variation. In locis of rs691870665 and rs80969284, these values were all higher, and the genetic variation was rich. The results suggested that the overall purity of CYP2E1 gene in Congjiang xiang-pigs was high, which establish a good foundation for animal model of drug metabolism.
