摘　　要：目的:研究KIF20A对肝癌SMMC-7721细胞系增殖和侵袭迁移能力的影响。方法:利用RNA干扰技术下调肝癌细胞系SMCC-7721中KIF20A的表达。qRT-PCR和Western blot分别检测细胞KIF20A在mRNA和蛋白水平表达中的变化。CCK-8实验检测增殖能力的改变。Transwell实验检测迁移、侵袭能力的改变。结果:qRT-PCR和Western blot结果显示KIF20A-siRNA能够成功下调KIF20A的表达,CCK-8和Transwell实验分别证实肝癌SMMC-7721细胞增殖和侵袭迁移能力下降。结论:抑制KIF20A的表达能够降低肝癌细胞的增殖和侵袭迁移能力。Objective：To investigate the protective effects of KIF20A on regulation of proliferation,migration and invasion in human hepatocellular carcinoma（HCC）cell line SMCC - 7721. Methods：KIF20A was down - regulated by using small interfering RNA（siRNA）in SMMC - 7721 cells. The transfection efficiency was detected with qRT -PCR and Western blot assay. CCK - 8 assay and Transwell migration and invasion assays were performed to evaluate the proliferation,migration and invasion ability respectively. Results：KIF20A was significantly down - regulated in SMMC - 7721 cells. After down - regulation of KIF20A,the proliferation,migration and invasion ability of SMMC -7721 were impaired. Conclusion：Down - regulation of the expression of KIF20A could inhibit cell proliferation,inva-sion and metastasis ability of HCC.
