摘 要:以长白山胡桃楸种仁壳为原料,精制种仁壳多糖并研究其抗氧化作用。通过回流提取、醇沉、有机溶剂洗涤、脱蛋白、脱色、真空冷冻干燥得到精制的种仁壳多糖;采用分光光度法测定种仁壳多糖纯度,采用红外光谱法进行定性分析,并研究胡桃楸种仁壳多糖对ABTS、DPPH、羟自由基的清除作用和铜离子还原能力及总还原能力。结果表明,种仁壳多糖对ABTS、DPPH、羟自由基的清除作用和还原铜离子能力及总还原能力随质量浓度的增加而增加,呈现一定的剂量-效应关系,胡桃楸种仁壳多糖清除DPPH自由基作用明显强于BHT,对铜离子的还原能力明显强于Vc。结果表明胡桃楸种仁壳多糖具有明显的抗氧化作用。The polysaccharides from seed shell of Juglans mandshurica were refined and antioxidant effect was studied at Changbai mountain seed shell of Juglans mandshurica as raw material. The refined polysaccharides were got through reflux extraction, alcohol precipitation, organic solvent washing, deproteinization, decoloration and vacuum freeze-drying. The purity of polysaccharides of seed shell was determined by spectrophotometric method. The qualitative analysis of polysaccharides was carried out using infrared spectroscopy. And scavenging activities of seed shell polysaccharides on ABTS, DPPH, hydroxyl radical and copper ion reduction and total reducing power was investigated. The results showed that the scavenging activities and reducing power of polysaccharides from seed shell of Juglans mandshurica were strengthened with increasing of concentration, there were dose-effect relationship. And the scavenging activity of seed shell polysaccharides on DPPH free radicals was notably stronger than BHT, the reducing power of seed shell polysaccharides on copper ions reducing power was stronger than Vc. The polysaccharides from seed shell of Juglans mandshurica had significantly antioxidant effect.
