摘 要:目的探讨低机械指数超声辐照SonoVue对脑缺血模型大鼠脑组织细胞凋亡诱导的影响。方法 SPF级成年Wistar大鼠56只,随机分为空白对照组10只,低剂量模型组12只、中剂量模型组12只、高剂量模型组12只和高剂量造影剂组10只,其中对照组和高剂量造影剂组脑供血正常,低、中、高剂量模型组结扎双侧颈动脉制作脑缺血模型。5组大鼠均行颅顶开窗术人造声窗,应用Logiq E9彩超仪进行超声辐照,探头ML6-15,频率15MHz,机械指数0.09,软组织热指数0.00,辐照时间20min。辐照开始时,低、中、高剂量模型组和高剂量造影剂组分别经尾静脉快速推注0.18、3.6、9.0、9.0mL/kg SonoVue溶液。5组大鼠辐照结束后行组织病理学检查,观察脑组织形态学变化,采用TUNEL法检测各组脑细胞凋亡情况。结果低、中、高剂量模型组大鼠端脑顶叶、侧叶和海马区可见不同程度缺血病灶,呈局灶性和/或弥漫性缺血改变;对照组和高剂量造影剂组未见明显形态学改变;低、中、高剂量模型组脑细胞凋亡指数(488.30±133.73、416.96±109.63、423.97±100.38)明显高于对照组(78.94±39.38)和高剂量造影剂组(126.97±64.91)(P<0.01),低、中、高剂量模型组脑细胞凋亡指数比较差异无统计学意义(P>0.05),对照组脑细胞凋亡指数与高剂量造影剂组比较差异无统计学意义(P>0.05)。结论低机械指数为0.09条件下SonoVue微泡在给药剂量9.0mL/kg范围内对脑组织细胞凋亡无明显促进作用。Objective To investigate the effect of low mechanical index ultrasound irradiation with SonoVue on apoptosis of cerebral cells in cerebral ischemia rat models. Methods Fifty-six adult Wistar rats in SPF grade were randomly divided into blank control group (n= 10), low-dose model group (n= 12), medium-dose model group (n= 12), high-dose model group (n=12) and high-dose contrast agent group (n= 10). Blank control group and high-dose contrast agent group received normal blood supply, while low-, medium- and high-dose model groups were performed bilateral carotid ligation to establish cerebral ischemia models. All five groups were given calvarium fenestration to make artificial acoustic window. Logiq E9 color ultrasound was adopted for ultrasonic irradiation with the probe of ML6-15, frequency of 15 MHz, mechanical index of 0.09, thermal index of soft tissue of 0.00, and irradiation time of 20 min. At the beginning of irradiation, low-, medium- and high-dose model groups and high-dose contrast agent group were given 0.18, 3.6, 9.0 and 9.0 mL/kg SonoVue solution with bolus injection via caudal vein. Histopathological examination was carried out after irradiation to observe the brain morphological changes in 5 groups, and TUNEL was used to detect the brain cell apoptosis. Results Ischemia lesions of different degrees were seen in the parietal lobe and lateral lobe of telencephalon and in hippocampus in low-, medium- and high-dose model groups, presenting as focal and/or diffuse ischemia change. No obvious morphological changes were seen in blank control group and high-dose contrast agent group. Apoptosis index of brain cells was obviously higher in low-dose model group (488. 30 ±133. 73), medium-dose model group (416.96 ± 109.63) and high-close model group (423.97 ±100.38) than that in control group (78.94 ±39.38) and high-dose contrast agent group (126.97±64.91) (P〈0.01), and there was no significant difference in pair-wise comparison of low-, medium- and high-dose mo
