摘 要:目的观察TMPRSS4基因沉默在甲状腺癌WRO细胞体外侵袭能力的影响。方法构建慢病毒载体转染甲状腺癌WRO细胞,分为三组:空白对照组(正常WRO细胞),NC-ShRNA阴性对照组,TMPRSS4-ShRNA组。使用WesternBlot方法检测转染后细胞的TMPRSS4表达,确定转染效果。采用Transwell法观察细胞体外侵袭力的改变。结果WesternBlot结果显示转染组TMPRSS4蛋白表达显著低于阴性转染组及空白对照组。侵袭实验结果提示TMPRSS4基因沉默组相比于空白对照组和阴性对照组侵袭穿膜细胞数显著减少(P<0.05)。结论下调TMPRSS4表达可有效减少甲状腺癌WRO细胞的侵袭能力。Objective To investigate the inhibitory effect of silencing Transmembrane protease serine 4 (TMPRSS4) gene in invasive ability of WRO cells of thyroid carcinoma in vitro. Methods WRO cells were established through transfection and were divided into three groups: the blank control group (normal WRO cells), the NC-ShRNA negative control group, and the TMPRSS4-ShRNA group. The expression of TMPRSS4 was detected by Western Blot and the transfection efficiency was determined. The invasiveness of the cells was measured by Transwell method in vitro. Results Western blot analysis showed that the expression of TMPRSS4 protein in the transfected group was significantly lower than that in the negative control group and blank control group. The experiment showed that silencing TMPRSS4 gene significantly reduced the number of invasion cells (P〈0.05) comparing with the blank control group and negative control group. Conclusion Down-regulation of TMPRSS4 expression can effectively reduce the invasive ability of WRO cells in thyroid carcinoma.
