摘  要:目的研究杨桃根中的有效成分杨桃根环己二酮(DMDD)对高脂饮食诱导的肥胖胰岛素抵抗小鼠的影响。方法采用高脂饮食喂养小鼠12周建立肥胖胰岛素抵抗小鼠模型,随后灌胃给药4周,每周称体重、测空腹血糖(FBG),15周末进行糖耐量试验,16周末,取血,处死小鼠,测量体长、腹围,称量附睾脂肪;比色法测血清总胆固醇(TC)、甘油三酯(TG),ELISA法测血清游离脂肪酸(FFA),空腹胰岛素(FINS),计算胰岛素抵抗指数(IRI)及胰岛素敏感指数(ISI);测定肝脏丙二醛(MDA)的含量、超氧化物歧化酶(T-SOD)及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果 DMDD各剂量组小鼠体重、体长、腹围、附睾脂肪、FBG、TC、TG、FFA、FINS、IRI、MDA水平明显降低,ISI、T-SOD和GSH-Px活性明显升高;DMDD各剂量组各个时间点的血糖及AUC均明显降低。结论 DMDD具有抑制肥胖、改善胰岛素抵抗及抗氧化的作用。Objective To investigate the effects of 2 - dodecyl - 6 - methoxycyclohexa - 2,5 - diene - 1,4 - dione(DMDD) on obesity and insulin resistance mice induced by high fat diet. Methods Mice were fed with high fat diet to establish obesity and in- sulin resistance model. Drugs were intragastrieally administered to mice for 4weeks. Body weight and blood glucose (FBG) were detected weekly. At the end of the 16tb week, intraperitoneal glucose tolerance test was conducted. Subsequently, their eyeball were taken for blood, body length, abdominal girth, fat around epididymis were measured. The concentration of serum TC, TG, FFA and insulin were detected, insulin resistance index (IRI) and insulin sensitive index (ISI) were calculated. Superoxide dis- mutase (SOD) glutathione peroxidase (GSH-Px) and malonyldialdehyde (MDA) were measured. Results Compared with the model group, body weight,abdominal girth, FBG, FINS, TG, TG, FFA, IRI, MDA , fat around epididymal were lower ; ISI, the activity of SOD and GSH - PX were increased ; Compare with the model group, glucose at each time point and AUC of DMDD groups was significantly lower. Conclusion DMDD has the effect of inhibiting obesity, improving insulin resistance, anti - oxida- tion of C57BL/6J mice induced by high fat diet.
