摘  要:为研究苯并[α]芘(BaP)对翡翠贻贝(Perna viridis)胚胎D型期的毒理作用,在观察翡翠贻贝胚胎发育的基础上,对D型期胚胎进行不同浓度的BaP胁迫实验,同时采用单细胞凝胶电泳技术探讨了96h时的胚胎细胞损伤程度。结果表明,BaP暴露对翡翠贻贝D型面盘幼虫的变态发育存在显著影响,随着暴露浓度的增大,畸形率显著增高,并呈现明显的剂量-效应关系;同时,翡翠贻贝D型面盘幼虫的半数致死浓度(LC50)随时间的推移逐渐降低,最终LC50在38.88μg/L处趋于稳定,其安全质量浓度为3.71μg/L。不同浓度的BaP暴露均能引起D型期细胞DNA损伤,其最大损伤程度可达3级。BaP胁迫对D型期胚胎细胞的彗星尾长、尾距及彗星尾部DNA含量的影响有显著的剂量-效应关系,3个指标具有较好的一致性。In order to research the toxic effects of benzo(a)pyrene (BaP) on the D-shaped veliger larvae cell of green mussel Perna viridis, the cellular damage degree at 96 h were studied by comet assay. Meanwhile, the morphology of green mussel was ob- served during metamorphosis development after IMP exposure. The results show that IMP has significant toxic effect on the malformation of the D-shaped veliger larvae. With the increase of the BaP concentration, the deformity rate increases significantly. The median lethal concentration (LC50) decreases with the increase of time then it tends to be constant. The stable concentration is 38. 88 μg · L-1 and the safe concentration is 3.71μg · L-1. The DNA damage in D-shaped veliger larvae cell can be induced under the different mass concentration of IMP, and the maximum damage degree can reach to the third level. There has a significant dose-effect relationship between the toxic effects with IMP concentration, additionally, the comet tail length, tail length and DNA content in the tail of larvae cell, the three indexes have a good consistency.
