层粘连蛋白配体及其受体在大鼠星形细胞及挫伤脑细胞中表达差异

摘  要:目的:观察层粘连蛋白配体(LN)与层粘连蛋白受体(LN-R)mRNA在大鼠星形细胞(Astrocyte)和挫伤脑细胞(Cell of TBI)之间表达的差异,探讨术中是否应尽量保存脑组织。方法从SD大鼠大脑皮层中培养星形胶质细胞,同时培养脑挫伤细胞(取自SD大鼠挫伤脑组织)。分别提取细胞总RNA,采用RT-PCR方法,分析LN和LN-R mRNA在大鼠星形细胞和挫伤脑细胞中表达的差异。结果 RT-PCT检测,大鼠星形细胞组和挫伤细胞组中均表达LN与LN-R mRNA,而挫伤脑细胞组中表达的LN和LN-R mRNA明显上升,差异有统计学意义。结论大鼠星形细胞和挫伤脑细胞中LN及LN-R表达存在明显差异,考虑与诱导神经干细胞迁移和分化等有关。重型颅脑外伤手术中,在无明显出血的情况下,应尽量保留挫伤脑组织。Objective To investigate the mRNA differential expression of Laminin (LN) and its receptor LN-R between rat astrocytes and cells around the injured cortex area after TBI and explore the necessity of preservation of brain tissue around the injured cortex on operation. Method Isolated and cultured rat astrocytes and cells around the injured cortex area after TBI respectively. Total RNA was extracted. Semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR) method was used in detection of the expression change of LN and LN-R between rat astrocytes and cells around the injured cortex area after TBI. Results LN and LN-R mRNA were expressed in rat astrocytes and cells around the injured cortex area after TBI. The mRNA expression level of LN and LN-R of rat cells around the injured cortex area after TBI were higher than these of rat astrocytes,the difference was significant,P〈0.05. Conclusion LN and LN-R differentially expressed in rat astrocytes and cells around the injured cortex area after TBI. We speculated this phenomenon was involved in biological process of the migration and differentiation of neural stem cells. During the operation of severe craniocerebral trauma,in the absence of obvious hemorrhage,brain tissue should be reserved as much as possible.
