摘　　要：目的研究OPN基因高表达的重组腺病毒对口腔鳞状细胞癌细胞恶性生物学行为的影响。方法培养口腔鳞状细胞癌Tca8113细胞株并将其分为rAd-OPN组和rAd-NC组,rAd-OPN组转染OPN基因高表达的重组腺病毒、rAd-NC组转染包含无效片段的重组腺病毒,测定细胞中OPN基因的m RNA含量以及细胞迁移、侵袭、上皮间质转化情况。结果腺病毒转染后12、24、36、48小时,rAd-OPN组细胞中OPN基因的m RNA含量以及细胞迁移率均显著高于rAd-NC组(P<0.05),穿膜细胞数显著多于rAd-NC组(P<0.05);腺病毒转染后48小时,rAd-OPN组中Twist、Snail、Vimentin、N-cadherin基因的mRNA含量显著高于rAd-NC组(P<0.05),E-cadherin基因的mRNA含量显著低于rAd-NC组(P<0.05)。结论 OPN基因高表达的重组腺病毒能够促进口腔鳞状细胞癌的迁移、侵袭以及上皮间质转化。Objective To study the effect of recombinant adenovirus with high expressed OPN gene on malignant biological behavior of oral squamous cell carcinoma cells. Method Oral squamous cell carcinoma cell line Tca8113 were cultured and divided into rAd-OPN group and rAd-NC group, rAd-OPN group was transfected with recombinant adenovirus with high expressed OPN gene; rAd-NC group was transfected with recombinant adenovirus containing invalid fragment. Then mRNA contents of OPN gene and migration, invasion, epithelial-mesenchymal of transfected cells were determined. Result 12, 24, 36, 48 hours after transfection, mRNA concents of OPN gene and cell migration rate in rAd-OPN group were significantly higher than rAd-NC group （P 〈 0.05）, transmembrane cell number was significantly greater than that of rAd-NC group （P 〈 0.05）; 48 hours after transfection, mRNA contents of Twist, Snail, Vimentin, N-cadherin gene in rAd-OPN group were significantly higher than rAd-NC group （P 〈 0.05）, while mRNA content of E-cadherin gene was significantly lower than rAd-NC group （P 〈 0.05）. Conclusion Recombinant adenovirus with high expressed of OPN gene can promote the migration, invasion and epithelial mesenchymal transition of oral squamous cell carcinoma.
