新生期大鼠间断吸入七氟醚对幼年期行为学及海马ApoE4的影响

摘　　要：目的：探讨新生大鼠间断吸入七氟醚对幼年期行为学及海马ApoE4的影响。方法：SD大鼠12只随机分成两组：吸入组（S组）和对照组（C组），每组6只。S组于出生后第7、14、21天吸入2．6％的七氟醚和运载气体（1L／min O2＋1L／minAir）2h，C组于相同时间点吸入运载气体2h。幼年期（第32—37天）行MWM．免疫组织化学法检测海马ApoE4的表达。结果：两组大鼠MWM中逃避潜伏期，原平台象限滞留时间百分比、运动距离百分比、进入次数百分比相比较差异无统计学意义（P〉0．05）；海马ApoE4在CA1、CA3、DG各区相比较差异均无统计学意义（P〉0．05）。结论：新生期间断重复吸入七氟醚未见海马ApoE4表达异常，幼年期学习与记忆无变化．临床重复多次使用是安全的。Objective To investigate the effect of mutiplesevoflurane anesthesia in beheavioristics and the level of ApoE4 in rat＇s hippocampus. Methods Twelve neonatal SD rats were randomly divided into 2 groups （n = 6） ： Sev group and Con group. The rats in the Sev group were exposed to 2.6% ssevofluraneand carrier gas for 2 h in P7, P14, P21, while the group in the Con exposure to 1 L/min O2＋1 L/min air last 2 h. Rats of infancy were tested via MWM in P32-P37 after the water maze experiment detected the level of ApoE5 in rat＇s hippocampus by immunohistochemistry. Results Compared the Sev group with the Con group, no significant change was found in the escape latency, the residence time, movement distance （P 〉 0.05）, the number of entering into the platform area （P 〉 0.05） of the MWM and the level of ApoE4 in CA1 ,CA3,DG area of rat＇s hippocampus（P 〉 0.05）. Conclusion Repeated inhalation of 2.6% sevoflurane in neonatal rats will not change the level of ApoE4 in rat＇s bippocampns, and will not affect the function of learning and memory during the neonatal period. It is safe to apply to clinic repeatedly.
