黄芩苷对牙周炎大鼠牙龈组织及外周血中IL-1β表达的影响

摘　　要：目的采用大肠杆菌脂多糖诱导大鼠牙周炎模型,观察黄芩苷对牙周炎大鼠牙龈组织及外周血中白介素-1β(interleukin-1β,IL-1β)表达水平的影响。方法选用50只8周龄SD大鼠,随机分成5组,每组10只,A组为对照组,B组为牙周炎组,C1、C2、C3组为黄芩苷治疗组,治疗1周后处死大鼠采取其外周血及牙龈组织,采用微量样本多重蛋白定量技术(CBA)检测外周血中IL-1β含量,采用RT-PCR法检测大鼠牙龈组织中IL-1βmRNA的表达水平。结果 B组大鼠牙龈组织中IL-1βmRNA及外周血中IL-1β表达含量均明显高于A组(P<0.05),C1、C2、C3组大鼠牙龈组织中IL-1βmRNA和外周血中IL-1β表达含量均明显低于B组(P<0.05)。结论黄芩苷可以降低牙周炎大鼠牙龈组织中IL-1βmRNA和外周血中IL-1β表达水平,抑制脂多糖对大鼠牙周组织的损伤作用。Objective To observe the effects of baicalin on the expression levels of interleukin-1β in gingival tissues and peripheral blood of rats with periodontitis that is induced by colibacillus lipopolysaccharide. Methods Fifty SD rats（ 8-week age） were randomly divided into five groups： control group（ group A）,periodontitis group（ group B）,baicalin treatment groups（ group C1,C2,C3）,with 10 rats in each group. After 1-week treatment,all the rats were sacrificed and gingival tissues and peripheral blood specimens were taken,and the expression levels of IL-1β mRNA in peripheral blood of rats were measured by cytometric bead array（ CBA）,which in gingival tissues of rats were detected by reverse transcription polymerase chain reaction（ RT-PCR） technique. Results The expression levels of IL-1β mRNA in gingival tissues and peripheral blood of rats in group B were significantly higher than those in group A（ P〈0. 05）. Moreover the expression levels of IL-1β mRNA in gingival tissues and peripheral blood of rats in group C1,C2,C3 were significantly lower than those in group B（ P〈0. 05）. Conclusion The baicalin can decrease the expression levels of IL-1β mRNA in gingival tissues and peripheral blood of rats with periodontitis,and can improve the injury effect of lipopolysaccharide on gingival tissues of rats.
