摘  要:目的观察外源性β-arrestin2基因对人肺腺癌细胞A549增殖的影响。方法利用脂质体转染技术将真核表达重组体pc DNA3.1-β-arrestin2质粒和空载体pc DNA3.1质粒分别导入A549细胞,经G418筛选后获得稳定转染细胞克隆,RT-PCR和Western blotting检测β-arrestin2基因表达的影响。采用MTT(四甲基偶氮唑盐)法分析转染细胞的增殖情况。结果经脂质体转染和筛选,建立了稳定表达β-arrestin2基因的A549细胞系。与未转染组和转染空白载体组比较,转染β-arrestin2基因的细胞生长速度明显减慢(P<0.05)。结论β-arrestin2基因可能通过抑制P65活性而抑制人肺腺癌细胞A549的生长。Objective To observe the effects of exogenousβ-arrestin2 gene transfection on the proliferation of human pulmonary adenocarcinoma cells A549 in vitro.Methods The pcDNA3.1-β-arrestin2 plasmid and and empty carrier-pc DNA3.1 plasmid were introduced into A549 cells by liposomes transfection technique,then the cell clones that could express stably β-arrestin2 gene were established by G418 screening.The expression levels of β-arrestin2 were detected by RT-PCR and Western Blotting,respectively.The proliferation status of positive clones was observed by methythiazoletertraolium ( MTT) assay.Results The A549 cell line that could express stably β-arrestin2 gene was established by liposome transfection and screening.As compared with that in non-transfection group and blank carrier-transfection group,the growth velocity in the cells that were transfected byβ-arrestin2 gene was significantly decreased ( P 〈0.05).Conclusion The β-arrestin2 gene can inhibit the growth of of human pulmonary adenocarcinoma cells-A549 in vitro by inhibiting the activity of P65 protein.
