摘  要:目的:研究槐耳清膏抑制裸鼠胃癌腹膜转移的机制。方法:建立裸鼠胃癌模型24只,随机分为生理盐水组(n=12,每2天腹腔注射0.9%氯化钠溶液1.5m L)、槐耳清膏组(n=12,每2天腹腔注射15mg/m L的槐耳清膏1.5m L)。记录各组裸鼠体质量变化,实验性腹膜转移癌指数(e PCI),原位肿瘤相对体积(RTV),q RT-PCR检测各组胃癌组织PI3K、Akt、MMP-2、MMP-9、u PA m RNA表达情况。结果:生理盐水组与槐耳清膏组在体质量变化上无统计学差异,槐耳清膏组的实验性腹膜癌转移指数(e PCI)和RTV均显著低于生理盐水组(P<0.05),槐耳清膏组胃癌组织PI3K、Akt、MMP-2、MMP-9、u PA m RNA表达也低于生理盐水组(P<0.05)。结论:槐耳清膏可以抑制胃癌腹膜转移,可能是通过负性调控PI3K/Akt通路这一机制实现的。Objective: To study the mechanism of Huaier inhibiting peritoneal metastasis of gastric cancer in nude mouse. Methods: 24 nude mouse models of gastric cancer were established and randomly divided into normal saline group(n=12, intraperitoneal injection of normal saline 1.5mL every 2 days) and Huaierqing Paste group(n=12, intraperitoneal injection 15mg/mL of Huaier 1.5mL every 2 days). The indexes as body weight, experimental peritoneal carcinomatosis index(e PCI), and relative tumor volume(RTV) were recorded. The expression of PI3 K, Akt, MMP-2, MMP-9, and u PA m RNA in tumor tissue of nude mice in each group were determined by using q RT-PCR. Results: In terms of general status, Huaierqing Paste had no obvious effect on body weight compared to normal saline group. The e PCI and RTV of Huaierqing Paste group was obviously lower than that of normal saline group(P〈0.05), and the expression of PI3 K, Akt, MMP-2, MMP-9, and u PA m RNA in Huaierqing Paste group was also significantly lower than that of normal saline group(P〈0.05). Conclusion: Huaierqing Paste could effectively inhibit peritoneal metastasis of gastric cancer in nude mouse, which might be related with its negative regulation on PI3K/Akt signaling pathway.
