摘  要:目的探讨土槿皮乙酸(PAB)对人宫颈鳞癌Siha细胞的增殖抑制和凋亡诱导作用及其相关机制。方法根据不同PAB浓度和作用时间,将Siha细胞分为对照组(0μmol/L)及1、2、4、8、16μmol/L PAB组和24、48、72 h组。采用CCK-8试剂盒检测不同浓度(0、1、2、4、8、16μmol/L)PAB作用不同时间(24、48、72 h)后对Siha细胞的增殖抑制率。采用免疫印迹法检测不同浓度PAB(0、2、4、8μmol/L)作用48 h后Siha细胞PI3K/Akt/GSK-3β信号通路及凋亡相关蛋白Bax和Bcl-2表达变化。结果 1、2、4、8、16μmol/L PAB对Siha细胞增殖均有抑制作用,且呈剂量-时间依赖效应关系。与0μmol/L PAB对照组比较,2、4、8μmol/L PAB组Siha细胞PI3K、p-Akt、p-GSK-3β和抗凋亡蛋白Bcl-2蛋白表达明显下调,促凋亡蛋白Bax表达明显增加(P<0.05)。结论 PAB具有抗人宫颈鳞癌Siha细胞作用,且可能与PI3K/Akt/GSK-3β信号通路抑制及上调Bax和下调Bcl-2表达有关。Objective To explore the effect of pseudolaric acid B(PAB) on human cervical carcinoma cell line Siha and its mechanism. Methods Siha cells were divided into control group(0 μmol/L PAB) and 1, 2, 4, 8, 16 μmol/L PAB group according to the concentrations of PAB, and 24, 48, 72 h group according to the treating time of PAB. The inhibitory effect of different concentrations of PAB(0, 1, 2, 4, 8, 16 μmol/L) and different exposure time(24, 48, 72 h)on Siha cells were measured by CCK-8 essay. The protein expression levels of PI3 K, Akt, p-Akt, GSK-3β, p-GSK-3β, Bax and Bcl-2 were detected by immunoblotting. Results 1, 2, 4, 8, 16 μmol/L PAB had remarkable inhibiting effects on Siha cells and in a dose- and time-dependent manner. The protein expression levels of PI3 K, p-Akt, p-GSK-3β and Bcl-2 were decreased in 2, 4, 8 μmol/L group of Siha cells than 0 μmol/L group after 48 exposure(all P〈0.05). On the contrary, the protein expression level of Bax was increased(P〈0.05). Conclusion PAB can inhibit the proliferation of Siha cells, its mechanism may be related to the inhibition of PI3K/Akt/GSK-3β signaling pathway and the up-regulation of Bax and down-regulation of Bcl-2.
