摘  要:目的通过动物实验观察苏拉明复合黏弹剂对鼠眼后发性白内障预防的有效性,探讨其不良反应及安全剂量。方法选用健康大鼠制作白内障大鼠模型,制备不同浓度的苏拉明复合黏弹剂(500μg/ml、400μg/ml、300μg/ml、200μg/ml、100μg/ml、50μg/ml),行超声乳化白内障吸除术,术中应用普通黏弹剂及不同浓度的苏拉明复合黏弹剂,观察术后大鼠眼压、角膜混浊情况、前房反应及后囊膜混浊情况。结果不同浓度的苏拉明黏弹剂未对眼压产生明显影响,无明显前房炎症反应,所产生的前房炎症反应最重为房水闪辉(+),且最迟在术后第3天完全消失,差异无显著性(P>0.05);所有大鼠术后均出现切口部位的角膜轻度水肿,且在术后第3天消失,从角膜水肿消失的时间比较,差异无显著性(P>0.05);而后囊膜混浊情况,在浓度200μg/ml以上的苏拉明复合黏弹剂中均可阻止后囊膜混浊,且浓度>300μg/ml的苏拉明可更有效阻止后囊膜混浊(P<0.05),300μg/ml、400μg/ml和500μg/ml的苏拉明复合黏弹剂术后6个月后囊膜混浊情况比较,差异无显著性(P>0.05)。结论苏拉明复合黏弹剂能够有效预防后发性白内障,且安全不良反应少。Objective To investigate effectiveness,adverse reaction,and safe dose from suramin composite viscoelastics on rat eyes posterior capsular opacification(PCO) through animal experience.Method Healthy rats were chosen to make models of cataract,and prepare suramin composite viscoelastics with different concentration(500μg/ml、400μg/ml、300μg/ml、200μg/ml、100μg/ml、50μg/ml).Phacoemulsification is undergone.General viscoelastics and suramin composite viscoelastics with different concentration are applied in phacoemulsification.After the operation,rat intraocular pressure,opacitas corneae,anterior chamber reaction,and posterior capsular opacification are observed.Result Suramin composite viscoelastics with different concentration have not obvious influence on intraocular pressure.Anterior chamber inflammatory reaction is not obvious,and the heaviest anterior chamber reaction is aqueous flare(+),which will disappear on the third day after operation.As for the time of cornea edema disappearance,there is no significant difference(P〉0.05).For posterior capsular opacification,suramin composite viscoelastics with more than 200μg/ml concentration all can prevent it.What's more,suramin composite viscoelastics with more than 300μg/ml concentration can prevent more effectively posterior capsular opacification(P〈0.05).there are no obvious difference among 300μg/ml、400μg/ml、and 500μg/ml suramin after six months of surgery(P〉0.05).Conclusion This experience proves that suramin composite viscoelastic can prevent pco effectively,it is safe and with fewer adverse reactions.
