摘 要:目的观察局灶性脑缺血大鼠生长相关蛋白43(GAP-43)和微管相关蛋白2(MAP-2)的表达变化,及轴突生长抑制因子A(NogoA)拮抗剂NEP1-40给药后对其表达的影响,探讨NogoA抑制神经再生的可能机制。方法雄性SD大鼠48只,随机分为3组:假手术组、模型组和给药组,每组16只,采用线栓法建立大脑中动脉闭塞模型,给药组于脑缺血术后1d给予NEP1-40溶液侧脑室注射。术后1、3、7和14d用免疫组织化学法检测缺血侧纹状体区域GAP-43和MAP-2的表达变化。结果脑缺血后缺血侧纹状体区域,在模型组和给药组脑缺血后1、3和7d GAP-43阳性表达的吸光度值逐步增高,14dGAP-43表达有所下降,但均明显高于假手术组。给药组1、3、7和14dGAP-43阳性表达的吸光度值均明显高于模型组(0.19±0.18 vs 0.17±0.23,0.24±0.10 vs 0.21±0.04,0.30±0.21 vs 0.25±0.06和0.26±0.23 vs 0.23±0.08,P<0.05)。模型组和给药组各时间点MAP-2阳性表达的吸光度值逐渐升高,但均明显低于假手术组。给药组各时间点MAP-2阳性表达的吸光度值均明显高于模型组。结论抑制NogoA后可以通过增加缺血区GAP-43和MAP-2的表达,促进神经修复,这可能是NogoA抑制神经再生的机制。Objective To study the effect of NEP1-40 on expression of GAP-43 and MAP-2in rats following focal cerebral ischemia.Methods Forty-eight male SD rats were divided into sham operation group,MCAO model group and NEP1-40 treatment group(16in each group).A MCAO model was established by Longa occulsion.The rats in NEP1-40 treatment group received intracerebroventricular injection of NEP1-40 on day 1after operation.Expression of GAP-43 and MAP-2in ischemic striatum was detected with immunohistochemical staining on days 1,3,7and 14 respectively after operation.Results The expression level of GAP-43 gradualy increased in MCAO group and NEP1-40 treatment group on days 1,3and 7after operation and then decreased on day14 after operation and was always significantly higher than in sham group.The expression level of GAP-43 was significantly higher in NEP1-40 treatment group than in MCAO model group on days1,3,7and 14 after operation.The expression level of MAP-2was significantly higher in MCAO model group and NEP1-40 treatment group than in sham operation group and in NEP1-40 treatment group than in MCAO model group at different time points after operation(P〈0.05).Conclusion NogoA can upregulate the expression level of GAP-43 and MAP-2and can thus promote nerve repair possibly by inhibiting the nerve regeneration.
