摘  要:目的探讨miR-7对胃癌细胞SGC7901增殖、迁移及侵袭能力的影响及其与黏着斑激酶(FAK)相关性。方法培养人胃癌SGC7901细胞,将细胞分为miR-7mimics组、miR-阴性对照组和空白对照组,分别转染miR-7模拟物(miR-7mimics)、miR-阴性序列和不做任何处理。Real-time PCR检测miR-7表达,MTT法检测细胞增殖情况,Transwell小室检测细胞迁移能力和细胞侵袭能力,Western blot检测FAK和FAK Tyr397蛋白表达,利用Pearson相关分析对变量间相关关系进行分析。结果 miR-7mimics组细胞中miR-7相对表达量(2.86±0.47),显著高于miR-阴性对照组(1.03±0.15)和空白对照组(1.06±0.17),差异有统计学意义(F=635.985,P<0.01);miR-7mimics组24、48、72和96h细胞生长抑制率均高于miR-阴性对照组和空白对照组,差异有统计学意义(均P<0.05);迁移实验中,miR-7mimics组平均迁移细胞数[(159.52±5.94)/视野],显著低于miR-阴性对照组[(315.42±7.59/视野]和空白对照组[(307.53±8.16)/视野],差异有统计学意义(均P<0.05);侵袭实验中,miR-7 mimics组平均侵袭细胞数[(29.85±4.52)/视野],显著低于miR-阴性对照组[(85.42±5.59)/视野]和空白对照组[(88.53±6.16)/视野],差异有统计学意义(均P<0.05);miR-7mimics组细胞中FAK、FAK Tyr397蛋白表达量均低于miR-阴性对照组和空白对照组,差异有统计学意义(均P<0.05);Pearson相关分析显示,细胞中miR-7表达量与FAK和FAK Tyr397蛋白表达量均呈负相关(r=-0.724和-0.874,均P<0.01)。结论过表达miR-7可显著抑制胃癌SGC7901细胞增殖、迁移及侵袭过程,可能与负性调控FAK蛋白表达有关。Objective To investigate the effects of miR-7on the proliferation,migration and invasion of gastric cancer cell SGC7901 and its correlation with focal adhesion kinase(FAK).Methods Human gastric cancer SGC7901 cells were cultured and divided into miR-7mimics group,miR-negative control group and blank control group,in which cells were transfected with miR-7mimics,miR-negative sequences and given no treatment,respectively.The expression of miR-7was detected by using real-time PCR.The cell proliferation was tested by using MTT assay.The cell migration and cell invasion were detected by using Transwell chamber.The expression levels of FAK and FAK Tyr397 proteins were detected by using Western blotting.The correlations between the variables were analyzed by using Pearson correlation analysis.Results The expression level of miR-7in miR-7mimics group was(2.86±0.47),which was significantly higher than that in miR-negative control group and blank control group(F=635.985,P〈0.001),in which the expression levels of miR-7were(1.03±0.15)and(1.06±0.17),respectively.The cell growth inhibition rates were higher in miR-7 mimics group than in the negative control group and blank control group at 24,48,72 and 96h,respectively,with the differences being statistically significant(P〈0.05).In the migration experiment,the average number of migrating cells in miR-7mimics group was(159.52±5.94)per view,which was significantly lower than in miR-negative control group and blank control group[(315.42±7.59)per view and(307.53±8.16)per view,respectively](P〈0.05).In the invasive experiments,the average number of invasive cells was(29.85±4.52)per view in miR-7mimics group,which was significantly lower than in miR-negative control group and blank control group[(85.42±5.59)per view and(88.53±6.16)per view,respectively](P〈0.05).The expression levels of FAK and FAK Tyr397 proteins were lower in miR-7mimics group than in miR-negative control group and blank control group,and the d
