摘  要:目的评价板蓝根水提物体外抑制人H7N9禽流感病毒的药效。方法利用改良的方法提取纯化板蓝根水提物。在鸡胚尿囊腔中扩增人H7N9禽流感病毒,以药物毒性试验(MTT法)检测药物的毒性,在狗肾细胞上种植人流感病毒PR8株(A/PR/8/34,H1N1)和人H7N9禽流感病毒(A/Anhui/01/2013和A/Shanghai/01/2013),采用细胞病变抑制法(CPE法)在治疗作用策略下研究板蓝根水提物在体外抑制人H7N9禽流感病毒的药效。结果板蓝根水提物体外的半数有毒浓度(TC50)为>5 000μg/m L。板蓝根水提物在体外对于人流感病毒PR8株(A/PR/8/34,H1N1)的IC_(50)为2 500μg/m L,选择指数(SI)>2;对于人禽流感病毒H7N9(A/Anhui/01/2013)的IC_(50)为5 000μg/m L,SI>1;对于H7N9(A/Shanghai/01/2013)IC_(50)为2 500μg/m L,SI>2。板蓝根水提物在体外能抑制人H7N9禽流感病毒,且对于H7N9(A/Shanghai/01/2013)毒株的抑制效果更佳。结论板蓝根水提物在体外能有效抑制人H7N9禽流感病毒,药效和作用于人流感病毒药效相当。Objective It is to observe the inhibition effect of water extraction of radix isatidis on H7N9 avian influnza virus in vitro. Methods Purification water extraction of radix isatidis were extracted by modified method. Human HTN9 avian influnza viruses were amplificated in chick embryo allantoic cavity,and their toxicity was detected by MMT method. Human influenza virus PR8( A/PR/8/34, H1N1 ) and human H7N9 avian influnza virus( A/Anhui/01/2013, H7N9) and ( A/Shanghai/01/2013, HTN9) were planted in dog renal cells. Then the pharmacology of water extraction of radix isatidis in the inhibition of HTN9 avian influnza virus in vitro was studied by CPE method under strategy of the treatment. Results Half toxic concentration (TC50) of water extraction of radix isatidis in vitro was 5 000 μg/mL, IC50 of human influenza virus PR8 ( A/PR/8/34, H1N1 ) was 2 500 μg/mL, selective index (SI) 〉 2 ; ICso of human H7 N9 avian influnza virus ( A/Anhui/01/2013, H7N9 ) was 5 000 μg/mL, SI 〉 1 ; IC50 of human H7N9 avian influnza virus( A/Shanghai/01/2013, H7N9) was 5 000 μg/mL, SI 〉 1. Chemical analysis showed that the main component of water extraction of radix isatidis was polysaccharide, nucleoside compounds and many kinds of amino acid also could be found. Water extraction of radix isatidis could inhibit HTN9 avian influnza virus in vitro, and the inhibition for HTN9 avian influnza virus(A/Shanghai/01/2013, H7N9) was the best. Conclusion Water extraction of radix isatidis could effectively inhibit H7N9 avian influnza virus in vitro, and the effect was similar to that for human influenza virus.
