结肠癌根治术后血清VEGF、HIF-1α含量与肿瘤复发及复发病灶中癌细胞恶性程度的关系

摘　　要：目的：研究结肠癌根治术后血清血管内皮生长因子（VEGF）、低氧诱导因子-1α（HIF-1α）含量与肿瘤复发及复发病灶中癌细胞恶性程度的关系。方法：选择2012年8月-2013年5月在本院接受结肠癌根治术的134例患者进行3年随访,根据肿瘤复发情况分为复发组（n=34）和未复发组（n=95）,测定术后7d时血清中VEGF、HIF-1α的含量以及肿瘤复发病灶中原癌基因、抑癌基因的表达量。结果：复发组患者术后血清中VEGF、HIF-1α的含量显著高于未复发组（P〈0.05）;复发组中术后VEGF和HIF-1α高含量患者复发病灶中抑癌基因RNF181、p53、ASPP2、TSPYL5、PTEN的mRNA含量显著低于VEGF和HIF-1α低含量患者（P〈0.05）,原癌基因Survivin、hTERT、LTEM1、Hes1的mRNA含量显著高于VEGF和HIF-1α低含量患者（P〈0.05）。结论：结肠癌根治术后血清VEGF、HIF-1α含量升高与术后复发有关,血清VEGF、HIF-1α含量越高,癌细胞恶性程度越高。Objective：To study the relationship between serum VEGF,HIF-1αcontent and tumor recurrence as well as the malignant degree of cancer cells in recurrent lesions after radical operation for colon cancer.Methods：Follow up was conducted on a total of 134 patients who received radical operation for colon cancer in our hospital for three years and these patients were divided into the recurrence group（n=34）and non-recurrence group（n=95）according to tumor recurrence,and the VEGF and HIF-1αcontent in serum as well as the expression levels of proto-oncogenes and tumor suppressor genes in recurrent tumor lesions were detected on the 7th day after operation.Results：VEGF and HIF-1αcontent in serum of recurrence group after operation were significantly higher than those of non-recurrence group;tumor suppressor genes RNF181,p53,ASPP2,TSPYL5 and PTEN mRNA content in recurrent lesions of recurrence group of patients with high VEGF and HIF-1αcontent were significantly lower than those of patients with low VEGF and HIF-1αcontent,and proto-oncogenes Survivin,hTERT,LTEM1 and Hes1mRNA content were significantly higher than those of patients with low VEGF and HIF-1αcontent.Conclusion：In-creased VEGF and HIF-1αcontent in serum after radical operation for colon cancer are associated with postoperative recurrence,and the higher the VEGF and HIF-1αcontent in serum,the higher the malignant degree of cancer cells.
