摘  要:目的:研究壳聚糖(CS)/海藻酸钠(ALG)自组装表面涂层修饰后的PCL/辛伐他汀(SIM)电纺支架的性能及其对牙髓干细胞增殖的影响。方法:静电纺丝法制备PCL/SIM基膜,自组装技术对其表面进行CS/ALG涂层修饰。用接触角测量仪测量支架的亲水性、SEM观察支架的纤维形貌、紫外分光光度法检测辛伐他汀的累计释放量、CCK-8法检测支架浸提液对大鼠牙髓干细胞(r DPSCs)增殖的影响,并通过体外细胞粘附实验,观察r DPSCs在支架上的粘附生长状况。结果:涂层修饰后的电纺支架纤维表面粗糙,相互交错呈微孔结构,表明CS/ALG已组装到纤维表面;涂层修饰后的支架表面呈亲水性;PCL/SIM支架中的辛伐他汀呈低剂量平稳缓释趋势;PCL和PCL/SIM两种支架对r DPSCs的增殖均无明显影响(P>0.05);经涂层修饰后的支架更利于r DPSCs的粘附、增殖,并能促进细胞向支架内部迁移。结论:CS/ALG涂层修饰后的PCL/SIM电纺支架具有良好的生物相容性和药物缓释效果,有望成为牙再生组织工程中的一种新型生物支架。AIM: To study the property of chitosan( CS) and alginate( ALG) self- assembly modified PCL / simvastatin( SIM) electrospun scaffold,and to evaluate its effects on rat dental pulp stem cells( DPSCs).METHODS: PCL and PCL / SIM fiber membranes were prepared by electrospinning process. The fiber membranes were modified with CS and ALG by self- assemble method. Water contact angles on modified and unmodified PCL and PCL / SIM fiber film were measured by contact angle measurement. The morphology of the fiber films was observed by SEM. The drug release of simvastatin in PBS solution was detected by UV-visible spectroscopy. The rat DPSCs were cultured with the scaffold leaching liquor,the cell proliferation was measured by CCK-8 assay and the morphology of the cells was observed by SEM. RESULTS: The water contact angles of 180° were acquired after the self- assembly of the films. The PCL and PCL / SIM nanofibers had porous and interwoven structure,CS / ALG was successfully coated on the surface of electrospun fiber film. The PCL / SIM electrospun scaffold exhibited steady and long lasting drug release capacity with under lethal dose. The SEM results of rat DPSCs cultured on the scaffolds indicated that the modified fiber film had better compatibility than the unmodified scallolds. CONCLUSION: The electrospun PCL / SIM fiber modified with CS / ALG by self-assembly have good biocompatibility and great controlled-drug-release capacity.
