摘 要:目的通过HPLC法测定明日叶Angelica keiskei Koidzumi中异补骨脂二氢黄酮、异补骨脂查尔酮、异虎耳草素、北美芹素和花椒毒素的含有量,固相微萃取(SPME)-GC-MS法分析其中挥发性成分。方法该植物甲醇提取液的HPLC分析采用GL Sciences色谱柱(5μm,4.6 mm×250 mm);流动相乙腈-水,梯度洗脱;检测波长314 nm;体积流量1.0 m L/min;柱温28℃。GC-MS分析采用Agilent 19091S-433:93.92873 HP-5MS 5%Phenyl Methyl silox-60℃-325℃(325℃)色谱柱(30 m×250μm×0.25μm);柱温50℃;进样口温度250℃;载气体积流量1.0 m L/min;扫描范围15~500 amu。结果 5种成分在各自线性范围内线性关系良好(r≥0.997 2),平均加样回收率为97.78%~102.25%(RSD〈2.25%)。明日叶中有11种挥发性成分,9种是萜烯类,总相对含有量达96.45%。结论该方法准确可靠,可为明日叶的质量控制提供参考。AIM To simultaneously determine the contents of isobavachin,isobavachalcone,sopimpinellin,pteryxin and xanthotoxin in Angelica keiskei Koidzumi by HPLC and to analysis the volatile components by solid phase microextraction( SPME)-GC-MS. METHODS The HPLC analysis of methanol extract of this plant was performed on a 28 ℃ thermostatic GL Sciences column( 5 μm,4. 6 mm × 250 mm),with the mobile phase comprising of acetonitrile-water at a flow rate of 1. 0 m L / min in a gradient elution manner,and the detection wavelength was set at 314 nm. The GC-MS analysis was carried out on an Agilent 19091S-433: 93. 92873 HP-5MS 5% Phenyl Methyl silox-60 ℃-325 ℃( 325 ℃) column( 30 m × 250 μm × 0. 25 μm) at a carrier gas flow rate of1. 0 m L / min,the column temperature was set at 50 ℃,the inlet temperature was set at 250 ℃,and the scan range was 15-500 amu. RESULTS The five constituents showed good linear relationships within their own ranges( r≥0. 997 2),whose average recoveries were 97. 78%-102. 25%( RSD 〈 2. 25%). There were eleven vola-tile components in A. keiskei,nine of which were terpenes with the total relative content of 96. 45%. CONCLUSION This accurate and reliable method can be used for the quality control of A. keiskei.
