摘  要:目的观察肝素结合表皮生长因子(heparin-binding epidermal growth falrtor like growth factor,HB-EGF)对部分肝移植大鼠肝细胞Cyclin D1表达的影响,并探讨其促进肝再生的机制。方法首先采用改良的"两袖套"法构建大鼠部分(60%)肝移植模型,共75只,受体随机平均分为A、B、C三组。其中,A组经皮下注射生理盐水(100 mg/kg),B组皮下注射HGF(100 mg/kg),C组为皮下注射HB-EGF(100 mg/kg),1次/d。手术后第1天、第2天、第3天、第5天和第7天各处死5只受体大鼠,采用实时荧光定量PCR、Western blotting检测肝组织中Cyclin D1 mRNA及蛋白的表达。结果 RT-PCR检测结果表明,C组术后第1天Cyclin D1 mRNA表达就达到高峰,且显著高于A、B组(P<0.05),之后1周逐渐降低;Western blotting检测结果表明,三组Cyclin D1蛋白表达水平均有上升,但C组术后第1天即达到高峰,且显著高于A、B组(P<0.05)。结论 HE-EGF可能通过上调肝移植后肝细胞Cyclin D1的表达来促进肝再生。Objective To observe effect of using heparin-binding epidermal growth factor-like growth factor (HB- EGF) on the expression of CyclinD1 of hepatic cell after partial transplantation in rats so as to explore its mechanism. Methods The Two-cuff technique was adopted firstly to construct liver transplantation model of rat part (60%) , totally 75 rats. The receptors were divided averagely and randomly into 3 groups: group A, B and C. Then 100 mg/kg the 0.9% NaCl (group A), 100 mg/kg HGF (group B) and 100 mg/kg HB-EGF (group C) were administered, respectively. All groups were done once a day. Subsequently, 5 receptors rats died for each group on the 1st, 2nd, 3rd, 5th and 7th days after the operation. Real-time fluorescence quantitative PCR and Western blotting were used to detect ex- pressions of CyclinD1 mRNA and protein in the hepatic tissue. Results The expression of CyclinD1 mRNA in liver tissue peaked in group C, was higher than other groups on the 1st day after operation. The expression of CyclinD1 protein was increased in 3 groups, but that in the group C was higher than that in other two groups on the 1 st day. Conclusion HB-EGF can promote liver regeneration after partial liver transplantation in rats. The mechanism may be related with the increase of CyclinD1 mRNA and protein.
