摘 要:目的探究si RNA作用于不同类型子宫内膜癌细胞雌激素受体相关受体α(estrogen receptor-related receptorα,ERRα)的表达及细胞的凋亡情况。方法 si RNA处理RL952、AN3-CA、HEC-1A和HEC-1B细胞,以RL952、AN3-CA为Ⅰ型子宫内膜癌体外模型,HEC-1A、HEC-1B为Ⅱ型子宫内膜癌体外模型;采用荧光定量PCR法检测ERRαm RNA的表达,Western blot法检测ERRα蛋白的表达,流式细胞仪分别检测si RNA干扰后4株子宫内膜癌细胞的凋亡情况。结果 si RNA组较对照组ERRαm RNA表达水平显著下降(RL952:0.701±0.017 vs.1.165±0.019;AN3-CA:0.899±0.019 vs.1.362±0.007;HEC-1A:0.093±0.004 vs.0.541±0.038;HEC-1B:0.158±0.004 vs.0.356±0.001;P均<0.05)。各株细胞si RNA组与对照组ERRαm RNA的表达比较,差异有统计学意义(P<0.05),ERRαm RNA表达分别减少了39.77%、34.01%、82.82%和55.78%。West ern blot结果显示,4株细胞RNA干扰后ERRα蛋白表达与m RNA表达趋势一致。4株细胞凋亡率:RL952为(30.000±1.056)%vs.(10.572±1.027)%;AN3-CA为(32.931±1.613)%vs.(10.274±0.882)%;HEC-1A为(17.272±0.950)%vs.(8.704±0.608)%;HEC-1B为(18.703±1.877)%vs.(3.801±0.265)%,si RNA干扰4株细胞后凋亡率与各自对照组比较,差异均有统计学意义(P<0.05)。结论 si RNA能够有效抑制子宫内膜癌中ERRα的表达,ERRα低表达可能与子宫内膜癌细胞的凋亡增加密切相关。Objective To explore the expression of estrogen receptor-related receptor α ( ERR α ) mRNA interfered with siRNA and the apoptosis of different types endometrial cancer cells .Methods Detected RL952,AN3- CA,HEC-1A and HEC-1B cells mRNA and protein expression of ERR α by fluorescence quantitative PCR and Western blot respectively after siRNA. Then, the changes of cell apoptosis were measured by FCM. Results The mRNA expressions of ERR α in siRNA groups were significantly decreased than the control group, which were RL952 (0.701±0.017 vs 1.165 ±0.019), AN3-CA (0.899±0.019 vs 1.362±0.007), HEC-1A (0.093±0.004 vs 0.541±0.038) and HEC-1B (0.158±0.004 vs 0.356±0.001), respectively (P 〈 0.05) . While the expression levels of ERR α reduced 39.77%, 34.01% , 82.82% and 55.78% ,respectively. A similar result was observed in the protein expression of ERR α. FCM described the ratios of cell apoptosis in each cell line: RL952 (30.000±1.056)% vs (10.572±1.027)%,AN3-CA (32.931±1.613)% vs (10.274±0.882)%,HEC-1A (17.272±0.949)% vs (8.704±0.608)%,HEC-1B (18.703±1.877)% vs (3.801±0.265)%.The apoptosis ratio of CON group was outstanding lower than KD group in each cell (P 〈 0.05) . Conclusions siRNA can targeting inhibited the expression of ERR α in different types endometrial cancer cells; ERR α low expressing had a close relationship with the increasing cell apoptosis in endometrial cancer ceils.
