复方丹参注射液对大肠埃希菌致小鼠睾丸细胞凋亡的作用

摘　　要：目的建立雄性小鼠生殖系统的大肠埃希菌逆行性感染动物模型,探讨复方丹参注射液对该模型中睾丸细胞凋亡的抑制作用。方法建立雄性小鼠生殖系统的大肠埃希菌逆行性感染动物模型后,腹腔注射复方丹参注射液,7 d后观察小鼠的精子数量、精子成活率、精子畸形率的改变;用TUNEL方法检测睾丸组织的凋亡指数;用Western blot检测Fas和Fasl的表达水平。结果高剂量的复方丹参注射液,可明显升高大肠埃希菌生殖系统感染小鼠的精子数量、精子成活率,降低精子畸形率;而且睾丸的细胞凋亡指数、Fas和Fasl的表达水平也明显降低。结论复方丹参注射液可抑制大肠埃希菌生殖系统逆行感染后睾丸细胞的凋亡,并对睾丸的生精功能具有保护作用。Objective To investigate the inhibitory effect of compound salvia injection on apoptosis of testis induced by escherichia coli retrograde infection in mice. Methods After the model of retrograde infection of escherichia coli in genital system of mice was established, compound salvia injection was intraperitoneally injected. The amount of sperms, survival rate of sperms and rate of sperm deformation were observed on 7th day. Apoptotie index in testis was detected by TUNEL, and the expressions of Fas and Fasl was analyzed by Western blot. Results After the genital system of mice was infected by escherichia coli, compound salvia injection obviously increased the amount and survival rate of sperms, and re- duced the rate of sperm deformation, as well as decreased apoptotic index and the levels of Fas and Fasl. Conclusion Compound salvia injection can inhibit the apoptosis of testis induced by escherichia coli retrograde infection in mice, and protect the spermatogenic function of testis.
