摘 要:目的探讨Fli-1及其靶基因COL1A1、TNC在颅内动脉瘤患者血液单核细胞中的表达及作用。方法研究纳入41例颅内动脉瘤患者,其中25例动脉瘤破裂患者,16例动脉瘤未破裂患者;取病人的外周血,10例正常人外周血作为对照,分离血液单核细胞,提取总RNA,根据之前生物信息预测结果,采用实时定量RT-PCR检测Fli-1及其靶基因(COL1A1、TNC)在疾病和健康人群中的表达差异。结果在检测的Fli-1及靶基因COL1A1和TNC中,在颅内动脉瘤中差异表达的基因有2个,分别为Fli-1和COL1A1。进一步分组发现,相比于未发生破裂的动脉瘤患者,这两个基因在破裂的颅内动脉瘤患者中的表达有显著升高。结论破裂与未破裂颅内动脉瘤患者之间Fli-1及COL1A1的表达水平差异可能与动脉瘤的破裂表现存在相关性。Objective To investigate the expression of Fli-1 and its target genes in patients with intracranial aneurysm. Methods Blood samples from 41 intracranial aneurysm patients( 25 ruptured and 16 unruptured) and 10 healthy controls were collected. Peripheral blood mononuclear cells( PBMCs) were isolated for total RNA exaction. The expression of 3 candidate genes predicted by informatics method were assessed by real-time quantitative PCR. Results Of the 4 candidate genes,2 differential expressed genes such as Fli-1and COL1A1 were found in patients with intracranial aneurysm. Further analysis revealed that the expression of Fli-1 and COL1A1 gene was significantly increased in patients with ruptured intracranial aneurysm compared with those in unruptured patients. Conclusion Differential expression of Fli-1 and COL1A1 between patients with and without ruptured intracranial aneurysm may be correlated with the ruptured phenotype in patients with intracranial aneurysm.
