摘 要:目的探讨Dicer与miR-155的表达在大肠癌发生发展中的临床意义。方法采用免疫组化法及实时荧光定量PCR法,分别检测大肠癌中Dicer与miR-155的表达,并分析二者的相关性,以及二者与大肠癌患者临床参数的相关性。结果与癌旁组织比较,大肠癌组织中,Dicer蛋白表达水平显著降低(P<0.01);与健康体检者比较,大肠癌患者miR-155表达水平显著增高(P<0.01)。与Dicer高表达患者比较,Dicer低表达患者外周血中miR-155表达水平显著增高(P<0.01)。与浸润深度及浆膜内者比较,浸润及浆膜外者Dicer表达水平显著降低(P<0.05),miR-155表达水平显著升高(P<0.01)。与Dukes分期A、B期比较,Dukes分期C、D期患者Dicer表达水平显著降低(P<0.05),miR-155表达水平显著升高(P<0.05)。结论大肠癌中存在Dicer表达降低以及miR-155表达增高,二者显著相关,且与大肠癌浸润和病程进展相关。Objective To investigate the expression and clinical significance of Dicer and miR-155 in the development of colorectal cancer. Methods The expression of Dicer and miR-155 in colorectal cancer were detected by immunohistochemistry and real-time fluorescence quantitative PCR,respectively,and the association of Dicer and miR-155 that with clinical parameters of colorectal cancer patients was analyzed. Results Compared with the adjacent tissues,Dicer protein expression level was significantly reduced in colorectal cancer tissue( P〈0. 01). Compared with healthy controls,miR-155 expression level in colorectal cancer patients were significantly increased( P〈0. 01). Compared with Dicer high expression of patients,Dicer miR-155 expression in peripheral blood of patients with low expression level was increased significantly( P〈0. 01). Compared with invasion of serosa inside,Dicer expression level was significantly reduced( P〈0. 05),and miR-155 expression level was significantly increased( P〈0. 01) in cancer tissue with invasion of serosa outside. Compared with Dukes stage A and B,Dicer expression levels were significantly reduced( P〈0. 05) and miR-155 expression level was significantly increased( P〈0. 05) in cancer tissue with Dukes stage C and D. Conclusion The expression of Dicer in colorectal cancer is reduced and the expression of miR-155 is increased,the two are significantly correlated,and is associated with the invasion and progression of colorectal cancer.
