摘  要:目的研究由缺血引起的脑血管疾病当中骨形态生成蛋白7(BMP-7)介导的脑缺血预处理(IPC)神经保护作用的细胞机制。方法采用原代皮层神经元建立神经元氧糖剥夺/复氧模型,用Western blot检测BMP-7及其下游信号的表达;用BMP抑制剂、Smad抑制剂及Smad 1 siRNA来研究BMP-7及其下游信号对氧糖剥夺预处理(OGDPC)中神经保护的作用。结果 OGDPC可诱导原代皮层神经元表达BMP-7,内源性和外源性的BMP-7都可以模拟OGDPC的神经保护作用。结论 Smad信号通路参与了OGDPC通过BMP-7发挥的神经保护作用。Objective To investigate the cellular mechanisms of bone morphogenetic protein 7( BMP-7) mediated neuroprotective effect of cerebral ischemic preconditioning( IPC) in the cerebral vascular diseases caused by ischemia. Methods The primary cortical neurons were used to establish the model of oxygen glucose deprivation / reperfusion( OGD / R). Western blot was used to detect the protein expression of BMP-7 and downstream signals. BMP inhibitor,Smad inhibitor and siRNA of Smad 1 were used to investigate the role of BMP-7 and its downstream signals in the neuroprotective effects of oxygen-glucose deprivation preconditioning( OGDPC). Results The results showed that OGDPC induced over expression of BMP-7 in neurons and SH-SY5 Y cells. Both of endogenous and exogenous BMP-7 could replicate the neuroprotective effects seen in OGDPC pretreatment. Conclusion Smad signaling pathway was found participated in the neuroprotective effects mediated by BMP-7 of OGDPC.
