摘 要:目的观察CD88在胃癌组织中的表达水平,探索CD88的表达水平对胃癌细胞侵袭能力的影响及其机制。方法采用实时定量聚合酶链反应(Real-time PCR)和Western blot方法检测CD88在癌及癌旁组织中mRNA和蛋白的表达水平。使用CD88特异性小干扰RNA(siRNA)转染人胃癌细胞株SGC7901,然后采用划痕及transwell方法检测细胞侵袭能力改变。结果在胃癌组织中,CD88 mRNA和蛋白表达水平明显比癌旁组织高,沉默CD88后,SGC7901的侵袭迁移能力明显减弱。结论 CD88在胃癌中表达增加,并可增强胃癌细胞的侵袭迁移能力,可为胃癌的靶向治疗研究提供新的靶点。Objective To study the expression level of CD88 in gastric cancer and normal tissue. To investigate the affection and mechanism of CD88 on migration and invasion activity of gastric cancer cell. Methods The different expression of CD88 protein levels in gastric carcinoma tissues and peritumoral tissues were determined by RT-PCR and Western blot respectively. After transfected with CD88 specific small interfering RNA( CD88-siRNA),the ability of migration and invasion of SGC7901 were examined by wound scratching and transwell assay. Results The CD88 mRNA expression was significantly higher in gastric carcinoma tissues than that in the peritumoral tissues. CD88-siRNA can obviously inhibit cell activity of invasion and migration of SGC7901. Conclusions The expression levels of CD88 was increased and significantly enhanced the ability of invasion and migration in gastric cancer cell. This study provide new target for the treatment of gastric cancer.
