TRIM22在宫颈癌中的表达及其与临床病理特征的关系

摘　　要：目的:探讨具有抗病毒和抗肿瘤潜能的三结构域分子22(TRIM22)在宫颈癌中的表达及其与临床病理特征的关系。方法:采用免疫组化SP法对38例宫颈癌组织、20例正常宫颈组织的TRIM22蛋白进行定位和定性分析;免疫蛋白印迹法(western blot)检测TRIM22蛋白表达情况。结果:(1)宫颈组织中TRIM22定位于细胞质和细胞核,宫颈癌组织中TRIM22的阳性率和染色强度明显低于正常宫颈组织(P<0.05);(2)宫颈癌组织中的TRIM22蛋白表达量低于正常宫颈组织(P<0.05),TRIM22的低表达与宫颈癌浸润深度、细胞学分级和淋巴结转移有关。结论:TRIM22低表达与宫颈癌恶性生长及不良预后有关,有望成为判断宫颈癌预后的标记物和基因辅助治疗的靶点。Objective：To investigate the expression of Tripartite Motif 22（TRIM22）which have antiviral and antitumor potentiality in the cervical tissues.Methods：20normal cervical tissues and 38 cervical cancer tissues were colleted for this study.The location and expression of TRIM22 were examined with immunohistochemistry and western blotting.Results：（1）TRIM22 mainly located at cytoplasm and nucleus;（2）the expression level of TRIM22 was significantly lower in cervical cancer tissues than that in normal cervical tissues（P〈0.05）.The decreased TRIM22 expression was associated with histological differentiation,invasive depth and lymph node metastasis（P〈0.05）.Conclusion：The results suggested that down-regulated TRIM22 expression is inversely associated with malignant potential or differentiation of cervical cancer,and it could be considered as a prognosis biomarker and therapeutic target.
