摘 要:目的 初步了解中卫地区宿主猪中小肠结肠炎耶尔森菌的携带情况、血清型、流行株型别和毒力特征,为该病的防治提供理论依据。方法 采用增菌后直接划线和PCR(聚合酶链反应)-直接划线对小肠结肠炎耶尔森菌分离培养,并对分离株进行血清分型、生物分型及毒力基因检测,同时对致病性菌株进行脉冲场凝胶电泳(PFGE)分型分析。结果 175份咽拭子标本经平板划线分离培养,共检出42株菌,检出率为24.00%;采用PCR-平板划线法检测共检出53株目标菌,检出率为30.29%;53株小肠结肠炎耶尔森菌共发现3种血清型,分别为O∶3、O∶5和O∶8;生物型为3型和1A型;毒力基因型别为35株“-+++++”型、9株“-++--+”、2株“+-----”型和7株“------”型;PFGE分型结果显示,所有试验菌株PFGE带型相似系数在80%~100%,共分成8种带型,以K6GN11C30021、K6GN11C30048、K6GN11C30012、K6GN11C300107为主要型别,占所有分型菌株的56.60%(30/53)。结论 PCR-平板划线分离培养方法明显优于平板划线法,且中卫市小肠结肠炎耶尔森菌血清型、生物型、毒力基因主要型别与宁夏总的监测结果一致。Objective To understand carrying case, serotype type and virulence characteristics in central region pig host en- terocolitis Yersinia enterocolitica,and to provide basis for the prevention and treatment of the disease. Results 175 samples of throat swab specimens were isolated and cultured by the plate, a total of 42 strains of bac- teria detection rate was 24.00%, by PCR detection fiat line method, a total of 53 strains detected target bacteria, detection rate is 30.29% ;53 strains enterocolitis Yalesen bacteria were respectively found three serotype, O: 3, O: 5 and O: 8 ; Biological type for type 3 and 1 atype;Virulencegenesdon'tfor35 strains"- + + + + +",9"- + + +" 2of" + ",type and 7 strains type ; PFGE classification, according to the results of all test strains PFGE with similarity coefficient between 80% - 100%, was divided into eight kinds of belt type, K6GN11 C30021, K6GNll C30048, K6GN11 C30012, K6GN11 C300107 don't, as the main type of all points type strains of 56.60% (30/53). Conclusion The PCR - tablet crossed flat line method is obviously better than the method of cultivation, and the enterocolitis Yalesen bacteria serotype, biological type, virulence genotype Zhongwei city consistent with the overall monitoring results in Ningxia.
