摘 要:目的探讨胃癌细胞中MET基因扩增与蛋白表达的相关性及其与临床病理特征的关系。方法应用荧光原位杂交(fluorescent in situ hybridization,FISH)及免疫组化Max Vision法同时检测85例胃癌石蜡包埋组织中MET基因扩增及蛋白表达。结果 FISH检测胃癌细胞中MET基因扩增率为5%(4/85);MET基因蛋白阳性率为43%(37/85);MET基因扩增与蛋白表达检出率不同,结果具有统计学意义(P<0.005),故两者关联性低。结论 MET基因扩增与蛋白表达不一致,原因有待进一步分析。Purpose To investigate the correlation of the MET gene amplification and protein expression in gastric cancer (GC) and its clinieopathological significance. Methods Fluorescent in situ hybridization (FISH) and immunohistoehemistry (IHC) of MaxVi- sion step were used to detect MET gene amplification and protein expression in paraffin-embedded tissues of GC. Result MET gene amplification rate was 5% (4/85) and its protein expression rate was 43 % ( 37/85 ). The rates of MET gene amplification and its pro- tein expression are different significantly (P 〈 0. 005). According to the statistical analysis, they had no correlation. Conclusion MET gene amplification is not consistent with protein expression, its underlying mechanism is to be determined.
