摘 要:油桐是我国重要的木本油料植物,过去对油桐的研究主要集中于栽培和常规育种,与油桐种仁油脂合成相关的分子机理研究还未见报道。本研究在油桐种仁油脂合成期的转录组数据基础上,通过生物信息学方法,将不饱和脂肪酸生物合成途径中的45条Unigene序列在KEGG(Kyoto Encyclopedia of Genes and Genomes)数据库中进行比对,获得了12个关键酶的同源蛋白质并对编码这些同源蛋白质的基因在油桐种仁油脂合成期的表达模式进行了分析。通过数据分析,本研究初步推导了油桐种仁不饱和脂肪酸合成途径,这将为油桐油脂合成,尤其是α-桐酸形成机理的解析奠定基础,并为进一步的理论研究和油桐的遗传改良提供潜在的基因资源。Tung oil tree(Vernicia fordii) is one of the important woody oil plants in China. Past researches on tung oil tree are mainly focused on the cultivation and conventional breeding while the molecular mechanisms related to tung oil synthesis are still uncovered. Based on the transcriptome data of tung tree kernel in oil synthesis stage, this research obtained twelve homologous proteins of key enzymes when the sequences of 45 Unigenes within unsaturated fatty acid biosynthesis pathway were aligned against KEGG(Kyoto Encyclopedia of Genes and Genomes) database and their expression profiles during oil synthesis stage of tung tree kernels were analyzed through bioinformatic methods. Through data analysis, this research preliminarily deduces unsaturated fatty acid biosynthesis pathway which not only shed light on elucidation of tung tree oil synthesis, especially α-eleostearic acid synthesis but also provides candidate genes for further theoretic researches and genetic improvement of tung tree.
