摘 要:目的:探讨化瘀通便汤对慢传输型便秘大鼠结肠水通道蛋白1(AQP1)和水通道蛋白3(AQP3)蛋白表达的影响。方法:72只雄性大鼠随机分为空白对照组,模型对照组,化瘀通便汤低、中、高剂量组,西药对照组,每组12只,采用大黄粉灌胃制造慢传输型便秘模型,计算各组大鼠碳末推进率,Western blot法检测大鼠近端结肠AQP1和AQP3蛋白(相对灰度值)的表达。结果:与空白对照组比较,模型对照组碳末推进率明显降低,大鼠结肠AQP1和AQP3表达升高,均有显著统计学意义(P<0.01);与模型对照组比较,化瘀通便汤中、高剂量组,西药对照组碳末推进率明显升高,各治疗组大鼠结肠AQP1和AQP3表达降低,均有统计学意义(P<0.05,P<0.01)。结论:慢传输型便秘大鼠便秘的发生可能与结肠组织中AQP1和AQP3的表达升高有关,化瘀通便汤治疗慢传输型便秘的机制可能通过降低结肠组织中AQP1和AQP3的表达来实现。Objective:To discuss the effects of Huayu Tongbian Decoction on slow transmit constipation(STC)mice colon in aquaporin1(AQP1)and aquaporin3(AQP3). Methods:72 male mice were randomly divided into blank control group,model control group,Huayu Tongbian Decoction of high,medium and low dose group,Western medicine control group(each group of 12). Using rhubarb powder to made STC model. To compute carbon propulsion rate and the protein expression of AQP1 and AQP3(relative grayscale values). Results:Compared with blank control group,model control group carbon propulsion rate significantly reduced and AQP1 and AQP3 expression increased,which had significant statistical significance(P〈0.01);Compared with model control group,Huayu Tongbian Decoction groups(high,medium)and Western medicine control group carbon propulsion rate increased significantly,eachtreatment group AQP1 and AQP3 expression reduced,which had statistical significance(P〈0.05,P〈0.01).Conclusion:Pathogenesis of STC may be related to the increase of AQP1 and AQP3 protein expression. The mechanism of Huayu Tongbian Decoction curing STC may be related to lower AQP1 and AQP3 protein expression.
