摘 要:目的:探讨Wnt信号通路蛋白在宫颈上皮内瘤变(CIN)和宫颈鳞癌中的表达及其与高危型HPV感染的相关性。方法:选取119例CIN或宫颈鳞癌患者为研究对象,并随机选取同期因子宫肌瘤行子宫全切术的37例正常宫颈标本作为对照组;利用实时荧光定量PCR和酶联免疫吸附试验分别检测Wnt-11、Frizzled/LRP和ROR2的基因和蛋白表达水平;应用HPV分型检测法测定高危型HPV感染状况。结果:与对照者相比,不同程度宫颈病变患者Wnt-11、LRP-6和ROR2基因和蛋白表达水平均明显升高(P<0.05),且从宫颈炎到宫颈鳞状细胞癌呈现逐渐增加的趋势(P<0.05);FZD-2在宫颈炎、CINⅠ-Ⅲ级的患者中均检测到轻微的增高,但组间比较差异未有统计学意义(P>0.05);共有62例检出高危型HPV,感染率为52.10%,高危型HPV感染率随着宫颈病变级别增高而呈现逐步递增(P<0.05);而且,高危型HPV感染患者的Wnt-11、LRP-6和ROR2表达水平明显高于无高危型HPV感染者(P<0.05);FZD-2的表达量则与是否HPV感染无明显相关(P>0.05)。结论:Wnt-11、LRP-6和ROR2在CIN和宫颈鳞癌中表达增强,且与高危型HPV感染呈正相关。Objective:To explore the expression of Wnt signaling pathway protein in cervical intraepithelial neoplasia and cervical squamous cell carcinoma as well as its correlation with the high-risk HPV infection.Methods:The 119 cases with cervical intraepithelial neoplasia and cervical squamous cell carcinoma were selected from January 2010 to June 2014.Another 37 cases of normal cervical specimens were chosen as the control group.The gene and protein expression of Wnt-11,Frizzled/LRP,and ROR2 in the Wnt signaling pathway were detected by real-time fluorescence quantitative PCR and ELISA.High-risk HPV infection was also determined by HPV genotyping.Results:Compared with controls,the levels of Wnt-11,LRP-6and ROR2 were significantly in-creased with different degree of cervical lesions(P〈0.05);A gradual increasing trend was also observed from the cervicitis to cervical squamous cell carcinoma(P〈0.05).Levels of FZD-2were slightly increased in the cervicitis,and CINⅠ-Ⅲ without statistical significance(P〈0.05).The total of 62 cases with high-risk HPV infection were detected,and the overall rate of highrisk HPV was 52.10%;High risk type of HPV infection rate was also gradually increased with degree of cervical lesions(P〈0.05);The expression levels of Wnt-11,LRP-6and ROR2 were significantly higher than those without high-risk HPV infection(P〈0.05);The expression of FZD-2had no significant correlation with HPV infection(P〉0.05).Conclusion:The gene and protein expressions of Wnt-11,LRP-6and ROR2 are increased in cervical intraepithelial neoplasia and cervical squamous cell carcinoma,showing positively correlated with the high-risk HPV infection.
