正常及脾阳虚大鼠脾组织蛋白组学差异分析

摘　　要：目的:通过比较正常及脾阳虚大鼠脾组织蛋白质谱,筛选和鉴定与脾阳虚证有关的疾病特异性蛋白,探寻脾阳虚证生物标志物。方法:选取SD大鼠20只随机分为对照组、脾阳虚组,每组10只。造模成功后无菌摘除脾脏,取出脾组织10mg/只,进行双向电泳实验、双向凝胶电泳图象分析,肽质量指纹图谱与电喷雾串联质谱鉴定蛋白质,分析差异蛋白质意义。结果:通过比较正常及脾阳虚证大鼠脾组织差异蛋白质,发现5个有意义的差异蛋白质:载脂蛋白A-I、真核翻译起始因子5a-1、3-磷酸甘油醛脱氢酶、结蛋白、异质核核糖核蛋白A2/B1。结论:正常及脾阳虚大鼠脾组织蛋白质存在差异表达。这些差异蛋白质可能是与脾阳虚证相关的疾病特异性蛋白,并有可能成为诊断脾阳虚证的分子标志物。Objective： To screen and identify the specific proteins related to syndrome of spleen yang deficiency by comparing protein profile of spleen tissues in normal and spleen yang deficiency rats, and to explore the biomarkers of syndrome of spleen yang deficiency. Methods： Twenty SD rats were randomly divided into the control group and the spleen yang deficiency group, ten rats in each group. After modeling, the spleen was removed by aseptic technic, 10 mg tissue of each spleen was analyzed by two-dimensional electrophoresis and two-dimensional gel electrophoresis image; the peptide mass fingerprinting and electrospray tandem mass spectrometry were used to identify and analyze the significance of differential proteins. Results： Five meaningful proteins were found by comparing differential proteins of spleen tissues between the normal and deficiency of spleen yang groups： loaded lipid apolipoprotein A-I, eukaryotic translation initiation factor 5a-1, glyceraldehyde 3-phosphate dehydrogenase, junction proteins, heterogeneous nuclear ribonucleoprotein A2/B1. Conclusion： There are differences of spleen protein expressions between the normal and spleen yang deficiency rats. These differential proteins may be the specific proteins that related to the diseases with syndrome of spleen yang deficiency, and may become molecular markers in the diagnosis of spleen yang deficiency syndrome.
