摘  要:目的:测定复序橐吾中游离和水解氨基酸的含量。方法:以异硫氰酸苯酯(PITC)为柱前衍生试剂,用反相高效液相色谱法(RP-HPLC)测定复序橐吾中氨基酸的含量。结果:21种氨基酸的检测浓度线性范围为0.0022-2.1000μmol/L,精密度RSD在0.38%-1.05%之间;稳定性RSD在0.46%-1.28%之间;游离氨基酸和水解氨基酸的重复性RSD在1.20%-2.38%和0.86%-2.43%之间;游离氨基酸和水解氨基酸的加样平均回收率分别在97.89%-101.05%和98.04%-103.03%之间且RSD均小于4%,游离和水解氨基酸的含量分别为0.2307%和8.7943%。结论:复序橐吾中含有21种氨基酸,并确定了其含量。Objective: To determinate the contents of free and hydrolysis amino acids in Ligularia Jaluensis. Methods: The amino acids contents in Ligularia Jaluensis were determined by using reversed phase high-performance liquid chromatography(RH-HPLC) with phenyl isothiocyanate(PITC) as the pre-column derivation reagent. Results: The 21 amino acids had good linearity in the range of 0.0022-2.1 000μmol/L. The RSD of precision was between 0.38% and 1.05%, the RSD of stability was between 0.46% and 1.28%, and the repeatability of free and hydrolysis amino acids was between 1.20% and 2.38%, or between 0.86% and 2.43%. The sample recovery rate of free and hydrolysis amino acids was between 97.89% and 101.05%, or between 98.04 % and 103.03%, and the RSD was less than 4%. The content of free amino acid and hydrolytic amino acid was 0.2307% and 8.7943%. Conclusion: There were 21 amino acids in Ligularia Jaluensis, and the contents of the 21 amino acids were determined.
