摘  要:为探讨生理性深层海水(PDSW)联合热疗对人胃癌SGC-7901细胞活力和凋亡的影响,通过培养人胃癌SGC-7901细胞株,随机分为实验组、对照组和空白组,实验组加PDSW,对照组加生理盐水,空白组不做其他处理,分别在37℃、40℃和43℃下培养,其中在40℃条件下培养6 h,43℃条件下培养1 h。用MTT法检测PDSW联合热疗在不同时间点的细胞活力,同时检测细胞克隆形成率,以及运用流式细胞分析检测细胞凋亡情况。结果表明,在热疗情况下,PDSW能抑制SGC-7901细胞的增殖,影响细胞克隆的形成;同时,PDSW联合热疗(40℃6 h和43℃1 h)后能显著增加SGC-7901细胞的凋亡。以上结果表明,PDSW联合热疗可使人胃癌细胞SGC-7901活力降低,增加胃癌细胞凋亡,具有抗肿瘤作用。To explore the effects of physiological deep sea water (PDSW) combined with hyperthermia therapy on activity and apoptosis of human gastric cancer cells SGC-7901, we cultured the gastric cancer SGC-7901 cells and randomly divided them into experimental group, control group and blank group. We added PDSW to the experimental group, added normal saline to the control group, and had no treatment on the blank group. We cultured the groups under 37℃, 40℃ and 43℃, respectively, and 40℃ for 6 h, 43℃ for 1 h. We used MTT method to detect the cell activity at different time points of PDSW combined with hyperthermia therapy. We also detected the cell colony formation rate, and detected the cell apoptosis by flow cytometry method. The results show that PDSW would inhibit the proliferation of SGC-7901 and affect the formation of cell clone combined with hyperthermia therapy. In addition, PDSW combined with hyperthermia therapy (40℃ 6 h and 43 ℃ 1 h) would significantly increase the apoptosis of SGC-7901 cells. These results showed that PDSW combined with hyperthermia therapy would decrease the activity and increase the apoptosis of human gastric cancer cells SGC-7901, and have anti-tumor effect.
