摘 要:【目的】评价束缚(immobilization,IMO)应激方法建立大鼠应激性心肌病(stress cardiomyopathy,SC)模型的有效性。【方法】健康雄性成年SD大鼠背部IMO 2 h,对照组麻醉后不束缚,检测二组大鼠心电图、左心室血流动力学、形态、血清去甲肾上腺素(norepinephrine,NE)及心肌损伤标志物(myocardial injury biomarker,cTnI)变化。【结果】大鼠IMO后5 min,与对照组比较:(1)心电图显示心率增加、ST段抬高、QTc间期延长。(2)血浆NE、cTnI浓度明显升高。(3)左心室内压、dp/dt明显升高。(4)超声心动图显示收缩期左心室心尖近球形扩张。【结论】大鼠IMO后迅速出现心电图、左心室血流动力学、左心室心尖扩张及血浆NE、cTnI浓度变化,符合SC主要临床特征。IMO应激模型建立方法简单,为SC基础研究提供了良好的动物模型。【Objective】To establish a rat model of stress cardiomyopathy(SC) induced by immobilization(IMO) and evaluate its effectiveness.【Methods】Healthy male adult SD rats were immobilized on their back to a board using adhesive tape for 2 h. Rats in control group were unrestrained after anesthesia. Characteristic changes in both groups were determined, such as cardiogram,hemodynamics of left ventricle, morphology and levels of serum norepinephrine(NE) and myocardial injury biomarker(cTn I).【Results】Compared with control group, the changes were observed 5 min after IMO including(1) remarkable tachycardia, rapid and persistent elevation of ST segment and prolonged QTc interval in cardiogram.(2) elevated concentrations of plasma NE and cTn I.(3)obvious elevation of left ventricle max pressure and dp/dt.(4) ballooning dilation of apical left ventricle in systole.【Conclusions】Changes of cardiogram, hemodynamics of left ventricle, dilation of apical left ventricle and concentrations of NE and cTn I occurred rapidly after rats were immobilized. These were in accordance with main clinical characteristics of SC. IMO stress model is established simply and potentially provides an experimental approach for basic research of SC.
