摘 要:目的探讨miR-133b及Bcl-w在甲状腺乳头状癌组织中的表达,以及抑制或过表达miR-133b对甲状腺乳头状癌细胞增殖和侵袭的影响。方法 Real-time PCR法检测17例患者甲状腺乳头状癌组织及对应的癌旁组织中miR-133b的表达,Western bolt法检测Bcl-w蛋白的表达。将miR-133b inhibitors及miR-133b mimics转染至甲状腺乳头状癌K1细胞株,Western blot方法检测转染后Bcl-w蛋白的表达变化,CCK-8方法和克隆形成实验检测转染后K1细胞的增殖和侵袭情况。结果 miR-133b在甲状腺乳头状癌组织中的表达水平明显低于癌旁组织(P<0.01),而Bcl-w蛋白在甲状腺乳头状癌组织中的表达水平明显高于癌旁组织(P<0.01)。转染miR-133b inhibitors的K1细胞增殖侵袭能力与对照组相比显著升高,细胞内Bcl-w蛋白表达亦明显升高(P<0.01)。转染miR-133b mimics的K1细胞增殖侵袭能力与对照组相比显著降低,细胞内Bcl-w蛋白表达亦明显降低(P<0.01)。结论 miR-133b抑制甲状腺乳头状癌K1细胞的增殖和侵袭可能与下调Bcl-w的表达相关。Objective To study the expressions of miR-133b and Bcl-w in thyroid papillary cancer(PTC) tissues and the effect of inhibitors or mimics of miR-133b on proliferation and invasion of thyroid papillary cancer cells. Methods The expression of miR-133b in 17 cases of thyroid papillary cancer tissues and corresponding para-carcinoma tissues was detected by real-time PCR, and Western blot method was employed to detect the expression of Bcl-w protein. The proliferation and invasion ability of thyroid papillary cancer K1 cells was detected by CCK-8 method and clone formation experiments respectively after miR-206 inhibitors or miR-206 mimics was transfected. Results The expression level of miR-133b in thyroid papillary cancer tissues was significantly lower than that in para-carcinoma normal tissues (P〈0.01) , but the expression lebel of Bcl-w protein in thyroid papillary cancer tissues was significantly higher than that in para-carcinoma tissues (P〈O.01) . The proliferation and invasion abilities of K1 ceils and the expression level of Bcl-w were increased significantly after miR-133b inhibitors was transfected (P〈0.01) , but lower significantly after miR-133b mimics was transfected (P〈O.01) . Conclusion The inhibition of the proliferation and invasion abilities of K1 cells by MiR-133b might be related to down-regulating Bcl-w expression.
