摘　　要：目的：研究香烟烟雾提取物对肺腺癌 A549细胞中 S100A2表达的影响。方法用含3%香烟烟雾提取物的培养基处理肺腺癌 A549细胞，对照组培养基不含香烟烟雾提取物。分别收集培养1、2、3周的细胞，采用实时荧光定量 PCR 法和 Western-blot 法检测各组细胞中 S100A2 mRNA 和 S100A2蛋白的相对表达量。结果经香烟烟雾提取物处理1、2、3周后，肺腺癌 A549细胞中 S100A2 mRNA 表达量逐渐增加，各处理组细胞 S100A2 mRNA相对表达量分别是对照组的（1.729±0.067）、（2.420±0.106）、（3.368±0.105）倍，差异有统计学意义（ F =140.441，P =0.000）。各组 A549细胞中 S100A2蛋白相对表达量分别为0.352±0.007、0.525±0.009、0.590±0.013，与对照组的0.241±0.006相比，差异有统计学意义（F =1941.418，P =0.000）。结论经香烟烟雾提取物可促进 A549细胞中 S100A2表达增高，为 S100A2成为吸烟的肺腺癌患者筛查的标志物提供了实验室依据。Objective To study the effect of cigarette smoke extract on the expression of S100A2 in lung adenocarcinoma A549 cell. Methods Lung adenocarcinoma A549 cells were treated by medium containing 3% cigarette smoke extract,control group used medium without cigarette smoke extract. A549 cells cultured for 1,2,3 weeks were collected separately,and the expressions of S100A2 mRNA and S100A2 protein were measured by real-time PCR method and western-blot method separately. Results After the cigarette smoke extract treated for one week,two weeks and three weeks,the expression of S100A2 mRNA increased gradually,were（1. 729 ± 0. 067）times,（2. 420 ± 0. 106） times and（3. 368 ± 0. 105）times to the control group ,the difference was statistically significant（F =140. 441,P = 0. 000）. The increasing expression of S100A2 protein in treatment group was time-dependence,were 0. 352 ± 0. 007,0. 525 ± 0. 009 and 0. 590 ± 0. 013,compared with the control group（0. 241 ± 0. 006）,the difference was statistically significant（ F = 1941. 418,P = 0. 000）. Conclusions The cigarette smoke extract can promote increased expression of S100A2 in A549 cells, from this study provided laboratory evidence for S100A2 become a marker screening of smokers as patients with lung adenocarcinoma.
