摘  要:目的建立不同产地藤茶的HPLC指纹图谱与一测多评方法,为藤茶的质量控制提供实验依据。方法采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.5%甲酸水溶液进行梯度洗脱,检测波长为258、292、369 nm,柱温为50℃,体积流量为0.8 m L/min,并应用SPSS 19.0软件对所得数据进行聚类分析和主成分分析,对不同产地藤茶样品进行质量分类。结果建立了不同产地藤茶的HPLC指纹图谱,确定了24个共有峰,鉴定了3个主要共有峰,分别为二氢杨梅素、杨梅素和杨梅苷,并建立了对不同产地藤茶中二氢杨梅素、杨梅素和杨梅苷的一测多评方法。结论建立的结合指纹图谱、模式识别和一测多评法的质量控制模式可应用于藤茶的质量评价。Objective A new quality evaluation method using HPLC fingerprint coupled with quantitative analysis of multi-components by single marker method was developed for the quality control of Ampelopsis grossedentata. Methods HPLC analysis was performed on an Agilent Eclipse Plus C18 (250 mm × 4.6 mm, 5 μn) column using methanol-0.05% aqueous solution of phosphoric acid as mobile phase at flow rate of 0.8 mL/min. The column temperature was set at 50℃ and the detection wavelength was at 258, 292, and 369 nm. The quality analyses of samples from different origins were accomplished by cluster analysis and principal component analysis using SPSS 19.0 statistical software. Results The HPLC fingerprint ofA. grossedentata from different regions was established. Twenty-four common peaks were found in the HPLC fingerprint of 14 samples, three important common components were identified as dihydromyricetin, myricitrin and myricetin. Further, the quantitative analysis of multi-components by single marker method of these contents was established. Conclusion The method of HPLC fingerprint combined with quantitative analysis of multi-components by single marker method can be used for the quality control ofA. grossedentata.
