LRP-5在不同分化程度胃癌组织中的表达及其意义

摘　　要：通过检测低密度脂蛋白受体相关蛋白5(LRP-5)在不同分化程度胃癌中的表达差异及其与胃癌患者临床病理资料之间的关系,探讨LRP-5在胃癌发病过程中的作用.采用实时荧光定量PCR及免疫组化的方法,对比分析了26例高分化、35例中分化、48例低分化胃癌及15例正常胃组织中LRP-5基因在转录和翻译水平的表达情况.实时荧光定量PCR和免疫组化结果均显示:LRP-5在中分化胃癌、低分化胃癌组织中的表达量高于正常胃组织,差异有统计学意义(P<0.05),在高分化胃癌组织中与正常胃组织之间比较无明显差别(P>0.05).结合临床资料分析表明,LRP-5蛋白在伴有淋巴结转移的胃癌组织中表达高于淋巴结未转移胃癌组织(P<0.05),与胃癌患者年龄、性别因素无关.提示LRP-5与胃癌的发生、发展、转移和恶性程度相关,可作为胃癌的诊断、分级和预后的判断依据.To explore the effect of low density lipoprotein receptor related protein 5 （LRP-5） in the pathogenesis of gastric cancer and the correlation between the expression of LRP-5 and the clini- copathological features,the expression of LRP-5 was detected by immuno-histochemical technique and real-time PCR in differently differentiated gastric carcinoma tissues, from which 26 cases were well differentiated, 35 were moderately differentiated and 48 were poorly differentiated. The results showed that, the expression of LRP-5 in the moderately and poorly differentiated gastric adenocarci- noma tissues were significantly higher than that of normal gastric tissue （P〈0.05）. There was no significant difference between the expression of LRP-5 in the well differentiated gastric adenocarci- noma tissues and normal gastric tissues （P〉0.05）. Combined with clinical data, the expression of LRP-5 protein in gastric cancer with lymph node metastasis was higher than that in gastric cancer without lymph node metastasis （P〈0.05）,which was not related to the age and sex of patients. The results indicated that LRP-5 may play an important role in the development of gastric cancer, which could be used for the diagnosis,grading and prognosis of gastric cancer.
